目的 對產于我國甘肅省的中藥秦艽的3種基源植物秦艽、麻花秦艽與小秦艽進行RAPD分析,建立具有鑒別意義的DNA指紋圖譜.方法 采用RAPD分子標記對秦艽的基因組DNA進行PCR擴增,利用NTSYSpc 2.1和Popgene 3.2軟件分析3種秦艽間的親緣關系,構建樹系圖.結果 從80條RAPD引物中篩選出4條具有鑒別意義的多態(tài)性引物,在3種秦艽中共得到39個DNA條帶,其中共有條帶2條,多態(tài)條帶37條.據此獲得了能有效區(qū)別3種秦艽的多態(tài)性RAPD指紋譜.結論 甘肅產秦艽的3種基源植物遺傳分化明顯,RAPD方法能有效地將它們區(qū)分鑒別.
