[摘要]
目的 考察壓敏膠基質(zhì)處方對青蒿琥酯體外透皮吸收的影響。方法 以聚丙烯酸酯壓敏膠為基質(zhì)設計不同的處方,考察了各種添加劑對青蒿琥酯在聚丙烯酸酯壓敏膠基質(zhì)中結(jié)晶的影響;通過改良的Franz透皮擴散池測定青蒿琥酯貼劑中青蒿琥酯透過SD大鼠腹部皮膚的滲透速率,以研究不同壓敏膠和不同添加劑對經(jīng)皮滲透速率的影響。結(jié)果 聚丙烯酸樹脂EudragitEPO 和聚乙烯吡咯烷酮(PVP-K30)對青蒿琥酯結(jié)晶均有較強的抑制作用,能提高青蒿琥酯在壓敏膠中的載藥量。體外透皮試驗結(jié)果顯示,儲存6周后,添加Eudragit EPO、PVP-K30的青蒿琥酯貼劑中的青蒿琥酯的累計透過量比無添加劑貼劑的高。結(jié)論 上述研究結(jié)果表明,提高青蒿琥酯在基質(zhì)中的載藥量有利于其開發(fā)成臨床應用的貼劑。

[Abstract]
Objective To investigate the effect of prescription of Pressure Sensitive Adhesives on the percutaneous absorption of artesunate. Methods The effects of various additives on the crystallization of artesunate in polyacrylate adhesive matrix were investigated by the various of polyacrylate pressuresensitive adhesives formulation, and the effects of various additives and pressuresensitive adhesives on the permeation of artesunate across SD rat abdominal skin were studied by modified Franz mothern method. ResultsPolymethacrylates Eudragit EPO and Poly(vinyl pyrrolidone) PVP-K30 inhibited the crystallization of artesunate in polyacrylate adhesive matrix. The addition of Eudragit EPO and PVP-K30 increase the flux of artesunate with the polyacrylate adhesive matrix after a storage period of 6 weeks.Conclusion These observations indicate that improvement in thesolubility of artesunate is necessary to realize fully the clinically applicable transdermal delivery system for drug.
