[摘要]
目的Ziziphora clinopodioides不同提取物對(duì)大鼠急性心肌缺血、對(duì)乳鼠心肌細(xì)胞缺氧復(fù)氧損傷的影響。方法 通過(guò)體內(nèi)注射垂體后葉素造成大鼠急性心肌缺血模型,觀察急性心肌缺血大鼠Ⅱ?qū)?lián)心電圖J點(diǎn)的變化,以及試藥對(duì)超氧化物歧化酶(SOD)、乳酸脫氫酶(LDH)、丙二醛(MDA)的影響。體外培養(yǎng)原代SD乳鼠心肌細(xì)胞,建立心肌細(xì)胞缺氧復(fù)氧損傷模型,用MTT法測(cè)定藥物對(duì)心肌細(xì)胞存活率的影響,測(cè)定給藥后MDA的變化。 結(jié)果 芳香新塔花石油醚部位和氯仿部位0.3 g/kg顯著抑制注射垂體后葉素所致心電圖J點(diǎn)升高,對(duì)心率、SOD、LDH、MDA的影響不顯著;乙醇提取物和氯仿部位能明顯降低缺氧-復(fù)氧模型下心肌細(xì)胞培養(yǎng)液中的MDA濃度。 結(jié)論芳香新塔花具有潛在的心肌缺血保護(hù)作。

[Abstract]
Objective To study the protective effects of ethanol extract and isolated fraction from Ziziphoraclinopodioides Lam. on acute myocardial ischemia of rats and neonatal rat cardiomyocytes damaged by hypoxia/reoxygenation stress. Methods Acute myocardial ischemia of rats was induced by injection of pituitrin. Electrocardiogram (ECG) was recorded, and the levels of lactate dehydrogenase (LDH),super-oxide dismutase (SOD) and the malondialdehyde (MDA) in medium were also tested. The neonatal rat cardiomyocytes were isolated and cultured in vitro to establish hypoxia/reoxygenation injury model The viability of cardiomyocytes was measured by MTT method.Change of MDA content in medium were also determined.Results Petroleum ether fraction and chloroform fraction (0.3 g/kg) remarkably inhibited the raise of J point caused by pituitrin, and significantly decreased MDA content in the model of hypoxia/reoxygenation injury. Conclusion Z. clinopodioides has a potential protective effects on acute myocardial ischemia and neonatal cardiomyocyte damage induced by hypoxiao/reoxygenation stress.
