目的 建立用反相高效液相色譜（RP-HPLC）法測定家兔眼房水中頭孢匹胺鈉的方法。 方法 將給予頭孢匹胺鈉后抽取的兔眼房水經(jīng)去蛋白處理后，采用RP-HPLC分析。流動相為磷酸二氫鉀溶液(0.05 mol/L)-甲醇（73∶27），檢測波長254 nm，體積流量1.0 mL/min，柱溫30 ℃。結(jié)果 頭孢匹胺鈉在0.12～2.50 μg/mL線性關(guān)系良好（r＝0.999 ），平均回收率為（94.30±0.96）％，RSD＜4％。結(jié)論 本法準(zhǔn)確，重現(xiàn)性好，適用于眼房水中頭孢匹胺鈉的分析。

Objective To establish a RP-HPLC method to determine the Cefpiramide level in rabbit ocular aqueous.Methods The proteins in samples were removed by precipitation. The RPHPLC method was performed by using sodium dihydrogen phosphate (aqueous solution， 0.05 mol/L) and methanol in a ratio of 73∶27 as mobile phase at a flow rate of 1.0 mL/min. The wavelength of detection was 254 nm and columntemperature was at 30 ℃.Results The calibration curves were linear over the range of 0.12～2.5 μg/mL and with a correlation coefficient of 0.999 9. The average recovery was (94.30± 0.96)％ and the RSD less than 4%.Conclusion The HPLC method is accurate and reliable for the determination of ocular Cefpiramide in rabbit eyes.
