[摘要]
目的 觀察肺纖方對(duì)博來(lái)霉素造模的肺纖維化大鼠肺組織病理形態(tài)發(fā)展的影響。方法 144只健康Wistar大鼠隨機(jī)分成對(duì)照組,模型組,陽(yáng)性組,肺纖方高、中、低劑量組,每組各24只,按Szapiel等法建立模型,次日ig給藥,肺纖方高、中、低劑量(3.2、1.6、0.8 g/kg)組均為10 mL/kg藥液ig給藥,陽(yáng)性對(duì)照組用醋酸潑尼松混懸液(6 mg/kg)ig給藥,對(duì)照組與模型組ig等量生理鹽水,1次/d,給藥周期21 d。分別在造模7、14、21 d后取材,前后期各隨機(jī)殺檢10只,中期殺檢4只對(duì)照驗(yàn)證。結(jié)果 與模型對(duì)照組相比,肺纖方高劑量組在改善大鼠外觀、體質(zhì)量、肺系數(shù)、肺臟病理組織形態(tài)學(xué)等方面效果較好(P<0.05或P<0.01),中劑量組幾乎與陽(yáng)性對(duì)照組作用相當(dāng),而陽(yáng)性對(duì)照組先期作用好于治療組,但中后期時(shí)肺纖方高劑量組的療效明顯好于陽(yáng)性組,說(shuō)明肺纖方抗大鼠肺纖維化病變的長(zhǎng)期療效優(yōu)于陽(yáng)性組。結(jié)論 肺纖方是防治肺纖維化的一種有效方劑,對(duì)博萊霉素致大鼠肺損傷具有一定的保護(hù)作用,對(duì)肺纖維化具有干預(yù)作用,且呈現(xiàn)一定的量效關(guān)系。

Objective To observe the curative effect of Formula for Lung Fibrosis on pulmonary fibrosis of rat model induced by Bleocamicina.. Methods Healthy Wistar rats (144) were randomly divided into six groups: control group, model group, prednisone acetate (PA, positive control) group, high-, mid-, and low-dose Formula for Lung Fibrosis groups, and there were 24 rats in each group. The rat model was established by Szapiel’s method. On the next day, the three treatment groups were ig administrated with high-, mid-, and low-dose Formula for Lung Fibrosis (3.2, 1.6, and 0.8 g/kg, 10 mL/kg body weight); The rats in PA group were ig administrated with PA (6 mg/kg); And the rats of control group and model group were ig administrated with physiological saline at the same dose, once daily for 21 d. After 7, 14, and 21 days of model establishment, 10 rats, 4 rats, and 10 rats were randomly sacrificed, respectively, and the materials were drawn, observed, recorded, and analyzed. Results Compared with the model group, high-dose Formula for Lung Fibrosis showed the significant difference on improving rat appearance, body weight, lung coefficient, and pathological histomorphology (P < 0.05 or 0.01). Mid-dose Formula for Lung Fibrosis had the same effect as PA group almost, and PA group was better than treatment group in beginning stage but worse in intermediate and later stages. This directed that high-dose Formula for Lung Fibrosis had better effect than PA group markedly, and the long-term effect of Formula for Lung Fibrosis was superior to PA. Conclusion Formula for Lung Fibrosis is an effective formula in treating and preventing pulmonary fibrosis. It could intervene in the injury of rats caused by Bleocamicina, and show a dose-effect relationship.
