[摘要]
目的 應(yīng)用高速逆流色譜法分離制備異槲皮素。方法 對(duì)棉花花提取物用高速逆流色譜法一步分離純化,以醋酸乙酯–水(1∶1)為兩相溶劑體系,上相為固定相,下相為流動(dòng)相,主機(jī)轉(zhuǎn)速為800 r/min,體積流量2.0 mL/min,檢測(cè)波長(zhǎng)254 nm,采用波譜法對(duì)所得化合物進(jìn)行結(jié)構(gòu)鑒定,薄層色譜、高效液相色譜法測(cè)定產(chǎn)品的純度。結(jié)果 在該條件下經(jīng)一步分離可得到質(zhì)量分?jǐn)?shù)為99%的異槲皮素對(duì)照品。結(jié)論 該方法操作簡(jiǎn)便,適合于從棉花花提取物中分離制備異槲皮素對(duì)照品。

[Abstract]
Objective To establish a method for the isolation and preparation of isoquercetin by high speed counter-current chromatography (HSCCC). Methods The total flavonoids in Gossypii Flos extract was further separated by HSCCC with a two-phase solvent system composed of ethylacetate-water (1:1) stationary phase. Main engine speed was 850 r/min and the detection wavelength was 254 nm at a flow rate of 2.0 mL/min. The obtained fraction was identified by spectral analysis and the purity was analyzed by TLC and HPLC. Results Isoquercetin reference substance with the purity of 99% under this given situation was obtained. Conclusion The results indicate that HSCCC is a simple and rapid method for the separation and preparation of isoquercetin reference substance.
