[摘要]
目的 研究硬棘軟珊瑚醋酸乙酯部位的化學(xué)成分。方法 采用正、反相硅膠柱色譜以及半制備高效液相色譜進(jìn)行分離,并根據(jù)理化性質(zhì)和波譜數(shù)據(jù)鑒定化合物的結(jié)構(gòu)。結(jié)果 分離得到11個(gè)化合物,其中3個(gè)環(huán)丁烯內(nèi)酯類化合物,分別鑒定為(S)-13-(2-羥基-3,4-二甲基-5-氧代-2,5-二氫呋喃-2-基)十三烷酸(1)、(S)-11-(2-羥基-3,4-二甲基-5-氧代-2,5-二氫呋喃-2-基)十一烷酸(2)、羥基二氫博伏內(nèi)酯(3);1個(gè)嘌呤類化合物,鑒定為1,3-二甲基黃嘌呤(4);7個(gè)甾體類化合物,分別鑒定為孕甾-20-烯-3-酮(5)、孕甾-1,4,20-三烯-3-酮(6)、3β-羥基-孕甾-20-烯(7)、3β-羥基-孕甾-5,20-二烯(8)、20-羥基-孕甾-1,4-二烯-3-酮(9)、20-羥基-孕甾-1-烯-3-酮(10)、17β-羥基-雄甾-1-烯-3-酮(11)。結(jié)論 環(huán)丁烯內(nèi)酯類化合物是首次從該屬軟珊瑚中分離得到,其中化合物1和2為新化合物,而硬棘軟珊瑚醋酸乙酯部位的化學(xué)成分主要為甾體類化合物。

[Abstract]
Objective To investigate the chemical constituents in the ethyl acetate fraction from Scleronephthya sp.. Methods The compounds were isolated by column chromatography (silica gel, including reversed phase) and semi-prepative HPLC, and their structures were identified on the basis of physicochemical properties and spectroscopic data. Results Eleven compounds were separated, three of which were butenolides, identified as (S)-13-(2-hydroxy-3,4-dimethyl-5-oxo-2,5-dihydrofuran-2-yl) tridecanoic acid (1), (S)-11-(2-hydroxy-3,4-dimethyl-5-oxo-2,5-dihydrofuran-2-yl) undecanoic acid (2), and hydroxydihydrobovolide (3), one of which was purine, identified as 1,3-dimethylxanthine (4), seven of which were pregnane steroids, identified as pregna-20-en-3-one (5), pregna-1,4,20-trien-3-one (6), 3β-hydroxy-pregna-20-ene (7), 3β-hydroxy-pregna-5,20-diene (8), 20-hydroxy-pregna-1,4-dien- 3-one (9), 20-hydroxy-pregna-1-en-3-one (10), and 17β- hydroxy-androst-1-en-3-one (11). Conclusion Pregnane steroids are the main chemical constituents in ethyl acetate fraction of Scleronephthya sp., and compounds 1 and 2 are new compounds, while butenolides are obtained from Scleronephthya sp. for the first time.
