[摘要]
目的 建立茯苓皮中松苓新酸的HPLC測定方法。方法 采用高效液相色譜法,Lichrospher C18色譜柱(200 mm×4.6 mm,5 μm),流動相為乙腈–0.5%磷酸水溶液(80∶20),檢測波長為242 nm,體積流量為1.0 mL/min,柱溫為30 ℃,進樣量為10 μL。結(jié)果 松苓新酸在0.142 5~2.280 0 μg與峰面積呈良好的線性關(guān)系(r=0.999 3),平均回收率為101.83%,RSD值為1.45%(n=6)。結(jié)論 該方法操作簡便,穩(wěn)定可行,重復(fù)性好,可用于茯苓皮中松苓新酸的測定。

[Abstract]
Objective To establish an HPLC method for the determination of dehydrotrametenolic acid in the peels of Poria cocos. Methods HPLC was carried out on a Lichrospher C18 column (200 mm×4.6 mm, 5 μm) with acetonitrile-0.5% phosphoric acid (80∶20) as mobile phase. The detection wavelength was set at 242 nm. The injection volume was 10 μL at the flow rate of 1.0 mL/min. The column temperature was at 30 ℃. Results The calibration curve was linear over the range of 0.142 5—2.280 0 μg (r=0.999 3) for dehydrotrametenolic acid. The average recovery rate was 101.83% with RSD value at 1.45% (n=6). Conclusion The established method is rapid, sensitive, and accurate, and could be used for the determination of dehydrotrametenolic acid in the peels of P. cocos.
