目的 構(gòu)建寡肽轉(zhuǎn)運(yùn)蛋白(PEPT1)高表達(dá)的Caco-2細(xì)胞系,并用于α-常春藤皂苷纈氨酸酯吸收機(jī)制的研究.方法 0.2 nmol/L瘦素誘導(dǎo)Caco-2細(xì)胞7 d以增加細(xì)胞上PEPT1的表達(dá)量.通過在瘦素誘導(dǎo)Caco-2細(xì)胞上進(jìn)行吸收抑制實(shí)驗(yàn),研究α-常春藤皂苷纈氨酸酯的吸收機(jī)制.結(jié)果 相對于未經(jīng)誘導(dǎo)的Caco-2細(xì)胞,α-常春藤皂苷纈氨酸酯在誘導(dǎo)的Caco-2細(xì)胞上攝取量提高了5.33倍,提示PEPT1的表達(dá)量增加了.結(jié)論 α-常春藤皂苷纈氨酸酯的吸收是由PEPT1介導(dǎo)的.

[Abstract]
Objective To establish a Caco-2 system with highly-expressed peptide transporter (PEPT1), and to apply this system to the absorption of L-valyl-α-hederin. Methods Caco-2 cells were induced with 0.2 nmol/L leptin for 7 d to increase the PEPT1 expression. The uptake mechanism of L-valyl-α-hederin was studied by inhibition absorption in leptin-treated Caco-2 cells. Results The uptake of L-valyl-α-hederin was 5.33 times higher in leptin-treated Caco-2 cells than those in the untreated Caco-2 cells which suggested more expression of PEPT1. Conclusion The uptake of L-valyl-α-hederin is mediated by PEPT1.
