[摘要]
目的 觀察催產(chǎn)素對3T3-L1脂肪細(xì)胞糖脂代謝的影響.方法 3T3-L1前脂肪細(xì)胞體外培養(yǎng),并誘導(dǎo)其分化成熟為脂肪細(xì)胞.研究催產(chǎn)素對脂肪細(xì)胞葡萄糖消耗量以及三酰甘油、游離脂肪酸和甘油的影響.采用實(shí)時(shí)熒光定量PCR法檢測糖脂代謝相關(guān)基因GLUT-1、GLUT-4、ATGT、HSL的mRNA表達(dá).結(jié)果 與對照組比較,催產(chǎn)素20、50、100 μg/mL組葡萄糖消耗量有所增加,且表現(xiàn)出劑量相關(guān).催產(chǎn)素組較對照組的三酰甘油降低,而甘油和游離脂肪酸增高.催產(chǎn)素50 μg/mL組中脂代謝相關(guān)基因HSL表達(dá)明顯高于對照組,糖代謝相關(guān)基因GLUT-4 mRNA表達(dá)水平增加.結(jié)論 催產(chǎn)素處理可減少3T3-L1細(xì)胞脂質(zhì)合成、增加脂質(zhì)分解作用,并可明顯改善脂質(zhì)積聚.

[Abstract]
Objective To study the effect of oxytocin on glucose and lipid metabolism in 3T3-L1 adipocytes. Methods Preadipocytes from 3T3-L1 cell line were cultured in vitro and induced to differentiate to adipocytes. Mature adipocytes were treated with oxytocin. Glucose consumption, triglyceride, free fat acid, and glycerol levels were determined. The mRNA expression of differentiation marker genes such as GLUT-1, GLUT-4, ATGT, and HSL were evaluated by RT-PCR method. Results The glucose consumption in the oxytocin (20, 50, and 100 μg/mL) groups were increased with dose-dependent relationship compared with control group. Triglyceride in the oxytocin group was lower than that in control group, while glycerol and free fatty acid decreased. There was significant increase of expression levels of lipid metabolism related gene HSL and sugar metabolism related gene GLUT-4 mRNA in the oxytocin (50 μg/mL) group compared with control group. Conclusion Treatment of oxytocin may reduce 3T3-L1 cell lipid synthesis, increase lipid decomposition, and obviously improve lipid accumulation.
