[摘要]
目的 研究苦豆子多糖溶液的構(gòu)象特征以及在不同因素影響下的溶液行為。方法 采用碘-碘化鉀反應(yīng)觀察苦豆子多糖鏈的分支情況和剛果紅試驗(yàn)研究構(gòu)象特征;采用圓二色譜(CD)法研究在不同濃度、溫度和添加鈣離子等條件下苦豆子多糖構(gòu)象的變化。結(jié)果 碘-碘化鉀反應(yīng)結(jié)果顯示苦豆子多糖有較長的側(cè)鏈和較多的分支;剛果紅試驗(yàn)中苦豆子多糖具有多股螺旋結(jié)構(gòu);濃度、溫度以及添加金屬離子使苦豆子多糖溶液構(gòu)象發(fā)生變化。結(jié)論 通過碘-碘化鉀反應(yīng)、剛果紅試驗(yàn)和圓二色譜法的結(jié)合可更加全面地了解苦豆子多糖溶液的構(gòu)象。

[Abstract]
Objective To study the conformation of Sophora alopecuroides polysaccharide (SAP) solution and its change under the condition of various factors. Methods Branch of SAP chain was observed by I2-KI reaction. And the conformation of SAP in solution was studied by Congo red experiment. The conformations of SAP in different conditions were analyzed by circular dichroism (CD) spectrum. Results I2-KI reaction showed that SAP had long chains and many branches. Congo red experiment showed that SAP was a multiple helical polysaccharide. The conformation of SAP solution can be affected by the changes of the concentration, temperature and metal ions. Conclusion The conformation of SAP solution can be studied more by methods of I2-KI reaction, Congo red experiment and CD spectrum.
