[摘要]
目的 考察毛葉疏花薔薇果提取物的亞慢性毒性.方法 選用96只健康SD大鼠隨機(jī)分為4組,雌雄各半,分別ig給予0、1.25、2.50、5.00 g/kg毛葉疏花薔薇果提取物,連續(xù)90 d,觀察動物生長情況.試驗(yàn)中、末期對動物的血液學(xué)和血清生化指標(biāo)進(jìn)行檢測,試驗(yàn)?zāi)┯嬎銊游镏饕K器的臟器系數(shù),并進(jìn)行組織病理學(xué)檢查.結(jié)果 試驗(yàn)期間動物無死亡.試驗(yàn)?zāi)└鲃┝拷M動物體質(zhì)量增長值、進(jìn)食量、食物利用率、臟器質(zhì)量、臟器系數(shù)與對照組比較差異均無統(tǒng)計學(xué)意義.在試驗(yàn)中、末期各劑量組動物血液學(xué)檢測指標(biāo)與對照組比較差異無統(tǒng)計學(xué)意義.血清生化檢測發(fā)現(xiàn),試驗(yàn)中期5.00 g/kg劑量組雌雄大鼠血清中的肌酐(CR)和雌性大鼠血清中的尿素氮(BUN)水平顯著升高;末期5.00 g/kg劑量組雄性大鼠血清中的CR水平仍居高不下,與對照組比較差異均有統(tǒng)計學(xué)意義(P< 0.05、0.01),而其他指標(biāo)均在正常值范圍內(nèi).病理組織學(xué)檢查發(fā)現(xiàn)5.00 g/kg劑量組部分動物腎小管和小腸可見相關(guān)病理改變.結(jié)論 在本實(shí)驗(yàn)條件下,5.00 g/kg毛葉疏花薔薇果提取物對SD大鼠的腎和小腸有損傷作用,最大無作用劑量為2.50 g/kg.

[Abstract]
Objective To study the subchronic toxicity of Rosa laxa var. mollis fruit extract. Methods SD rats (96) were divided randomly into four groups, and each group consisted of 12 males and 12 females. The animals in four groups were ig administered with R. laxa var. mollis fruit extract at doses of 0 (control), 1.25, 2.50, 5.00 g/kg for 90 d, respectively. During the entire course of the study, the rats were observed daily. The hematological and serum biochemical examinations were conducted after 16 h of starvation at 45th day as an intermittent study and 90th day as a terminal. All animals were calculated organ coefficients and studied for histological changes. Results At the end of the study, no deaths were observed in all groups. There were no differences in body weight gains, food intakes, food availability, and organ coefficients compared to the control (P> 0.05). For the hematological analyses, no parameters demonstrated significant changes in all groups during the entire study. During the serum biochemical analyses, there were significant increases of creatinine (CR) level in 5.00 g/kg both sexes and blood urea nitrogen (BUN) level in 5.00 g/kg female rats at the 45th day. Thereafter, significant increase of CR level in 5.00 g/kg male rats were observed at the end of the study (P <0.05, 0.01), but the other indexes were in the normal range. In 5.00 g/kg dose group, remarkable histopathological changes were observed in kidney and intestine of the SD rats. Conclusion R. laxa var. mollis fruit extract could be harmful to kidneys and intestines of the SD rats at 5.00 g/kg dose. No observed adverse effect level (NOAEL) is 2.50 g/kg.
