[摘要]
目的 考察清解方霧化吸入劑的抗炎作用及其作用機制。方法 采用二甲苯致小鼠耳廓腫脹實驗、冰醋酸致小鼠毛細血管通透性增加實驗以及角叉菜膠致小鼠足腫脹實驗,觀察清解方霧化吸入劑的抗炎作用。測定腫脹足部位炎癥因子NO、PGE2、IL-1β和TNF-α,探討其抗炎作用機制。結(jié)果 清解方霧化吸入劑9.2、18.4 g/kg均能顯著降低小鼠耳腫脹度(P<0.05、0.01),降低灌洗液吸光度值(P<0.05),抑制角叉菜膠引起的足跖腫脹(P<0.05),顯著抑制炎癥部位炎癥介質(zhì)NO、PGE2水平(P<0.01、0.001),顯著抑制炎癥部位細胞因子IL-1β、TNF-α的釋放(P<0.001),且清解方霧化吸入劑4.6 g/kg也顯著抑制IL-1β的釋放(P<0.01)。結(jié)論 清解方霧化吸入劑具有顯著的抗炎作用,作用機制可能在于降低IL-1β、TNF-α致炎細胞因子的釋放,減少炎癥部位NO、PGE2炎癥介質(zhì)的生成。

[Abstract]
Objective To investigate the anti-inflammatory effect of Qingjie Decoction Aerosol, and explore its mechanism. Methods Xylene induced ear edema experiment, glacial acetic acid increased capillary permeability experiment, and carrageenan induced foot swelling experiment were adopted to evaluate the anti-inflammatory efficacy of Qingjie Decoction Aerosol. Meanwhile, inflammatory factors NO, PGE2, IL-1β, and TNF-α in the swollen feet were determined to study its mechanism. Results Qingjie Decoction Aerosol (9.2, 18.4 g/kg) significantly reduced ear edema induced by xylene (P<0.05, 0.001), decreased absorbance values of lavage fluid (P<0.05), and inhibited carrageenin induced paw edema (P<0.05). Qingjie Decoction Aerosol (9.2, 18.4 g/kg) significantly inhibited levels of inflammatory mediators NO and PGE2 (P<0.01, 0.001), significantly inhibited realese of inflammatory cytokine IL-1β and TNF-α (P<0.001), Qingjie Decoction Aerosol (4.6 g/kg) also significantly inhibited the release of IL-1β (P<0.01). Conclusion Qingjie Decoction exhibited a prominent anti-inflammatory capacity, which might be mediate by IL-1β, TNF-α inhibition and NO, PGE2 reduction. Qingjie Decoction Aerosol has significant anti-inflammatory effect, which might be related to release reduction of proinflammatory cytokines IL-1β, TNF-α and generation reduction of inflammatory mediators NO and PGE2.
