[摘要]
目的 探討齊墩果酸對(duì)大鼠蛛網(wǎng)膜下腔出血后早期腦損傷的保護(hù)作用,闡釋其作用機(jī)制。方法 72只大鼠隨機(jī)分為假手術(shù)組、模型組和齊墩果酸10、20 mg/kg組,每組18只。采用頸內(nèi)動(dòng)脈穿刺法建立大鼠蛛網(wǎng)膜下腔出血模型,并在造模1 h后給藥,24 h后進(jìn)行神經(jīng)功能損傷評(píng)分;檢測(cè)腦水含率、腦伊文思藍(lán)浸出;并利用Western blotting法檢測(cè)腦組織VE-cadherin、p120-catenin蛋白的表達(dá)量。結(jié)果 齊墩果酸(20 mg/kg)可有效改善神經(jīng)功能損傷(P<0.01);降低腦水腫和腦血管通透性(P<0.01);增加VE-cadherin、p120-catenin蛋白的表達(dá)(P<0.01)。結(jié)論 齊墩果酸對(duì)大鼠蛛網(wǎng)膜下腔出血后早期腦損傷具有顯著的保護(hù)作用,改善腦水腫,其作用機(jī)制可能與增加腦血管內(nèi)皮粘附連接蛋白的表達(dá)有關(guān)。

[Abstract]
Objective To explore the effect of oleanolic acid on early brain injury after subarachnoid hemorrhage and explain its mechanism. Methods SD rats (72) were divided into 4 groups: Sham, model, and oleanolic acid groups (10 and 20 mg/kg), and each group had18 rats. The endovascular puncture model was used to induce subarachnoid hemorrhage. Rats were ig administered with the according drugs after 1 h of model established. Neurological score, brain water content and blood-brain barrier were evaluated after 24 h. The protein expression of VE-cadherin and p120-catenin were determined by Western blotting. Results Oleanolic acid (20 mg/kg) could significantly improve neurological scores (P < 0.01), reduced brain water content and blood-brain barrier permeability (P < 0.01), and increased the levels of VE-cadherin and p120-catenin (P < 0.01). Conclusion Oleanolic acid could protect against early brain injury after subarachnoid hemorrhage in rats and improve brain edema. The mechanism may be related to the increase of expression of endothelial adhesion protein.
