[摘要]
目的 觀察睡安片對(duì)咖啡因誘導(dǎo)失眠癥斑馬魚的鎮(zhèn)靜作用和睡眠改善作用,評(píng)價(jià)睡安片對(duì)失眠癥的療效。方法 采用200 μmol/L咖啡因誘導(dǎo)活體斑馬魚建立失眠癥模型,并確定睡安片的最大非致死濃度(MNLC)。睡安片選取MNLC(1 200 μg/mL)、1/3 MNLC(400 μg/mL)、1/10 MNLC(120 μg/mL)3個(gè)質(zhì)量濃度,同時(shí)設(shè)置對(duì)照組、模型組和氯化鋰(500 μg/mL)組,每組均處理30尾斑馬魚幼魚,使用行為分析儀測量斑馬魚的運(yùn)動(dòng)距離和處于失眠狀態(tài)的時(shí)間,計(jì)算鎮(zhèn)靜作用和睡眠改善作用。結(jié)果 睡安片的MNLC為1 200 μg/mL。120、400、1 200 μg/mL睡安片組與模型組比較,運(yùn)動(dòng)距離均顯著減少,差異具有極顯著性(P<0.001),鎮(zhèn)靜作用分別增加0.69、1.66、2.74倍。除120 μg/mL質(zhì)量濃度外,400、1 200 μg/mL組與模型組比較,失眠時(shí)間顯著減少,差異具有極顯著性(P<0.001),睡眠改善作用分別增加1.63、2.65倍。結(jié)論 睡安片對(duì)咖啡因誘導(dǎo)失眠癥斑馬魚具有明顯的鎮(zhèn)靜和睡眠改善作用,呈現(xiàn)明顯的濃度相關(guān)性。

[Abstract]
Objective To observe the sedative effect and sleep improvement of Shui'an Tablets on zebra fishes with insomnia caused by caffeine, and to evaluate the effect of Shui'an Tablets on insomnia. Methods The insomnia model was established by 200 μmol/L caffeine in living zebra fishes, and the maximum non-lethal concentration (MNLC) of Shui'an Tablets was determined. All zebra fishes were randomly divided into control group, model group, lithium chloride (500 μg/mL) group, and Shui'an Tablets 1/10 MNLC (120 μg/mL), 1/3 MNLC (400 μg/mL), and MNLC (1 200 μg/mL) groups, and each group had 30 zebra fishes. The distance of movement and the time of insomnia were measured with behavioral analyzer, and sedative effect and sleep improvement of Shui'an Tablets on zebra fishes was calculated. Results The maximum non-lethal concentration (MNLC) of Shui'an Tablets was 1 200 μg/mL. Compared with the model group, the exercise distances of Shui'an Tablets 120, 400, and 1 200 μg/mL groups were significantly decreased (P<0.001) and sedative effects were 0.69, 1.66, and 2.74 times, respectively. Compared with the model group, the times of insomnia of Shui'an Tablets 400 and 1 200 μg/mL groups were significantly decreased (P<0.001), and the improvements of sleep were0.02, 1.63, and 2.56 times, respectively. Conclusion Shui'an Tablets have obvious sedative and sleep improvement effects on zebra fishes with insomnia caused by caffeine, and show significant concentration-dependence.
