[摘要]
抗腫瘤抗生素來源于微生物的代謝物及其衍生物,是一類重要的細(xì)胞毒類抗腫瘤藥物,結(jié)構(gòu)獨(dú)特、活性優(yōu)良,但具有明顯的毒副作用也是其顯著的特點(diǎn)。近年來不斷有新的抗腫瘤抗生素被發(fā)現(xiàn),按照不同的作用機(jī)制,分別對(duì)干擾腫瘤細(xì)胞DNA結(jié)構(gòu)與功能,抑制腫瘤細(xì)胞分裂、增殖和遷移,抑制腫瘤細(xì)胞重要蛋白酶,誘導(dǎo)腫瘤細(xì)胞凋亡,通過自由基損傷發(fā)揮作用的抗腫瘤抗生素的研究進(jìn)展進(jìn)行綜述,并展望其在未來臨床中的應(yīng)用。

Antitumor antibiotics, derived from microbial metabolites and their derivatives, are an important class of cytotoxic antitumor drugs with unique structure and excellent activity, but their obvious toxic and side effects are also notable. New anticancer antibiotics have been found continuously in recent years. In this paper, antitumor antibiotics according to different mechanism of action such as interfering with the DNA structure and function of tumor cells, inhibiting tumor cell division, proliferation, and migration, inhibiting the important protease of tumor cells, inducing the apoptosis of tumor cell, acting through free radical damage are reviewed. And their future clinical applications are prospected.
