[摘要]
目的 探討dl-3-正丁基苯酞對內(nèi)皮素誘導(dǎo)腦缺血大鼠行為學(xué)的改善作用及其作用機(jī)制。方法 采用向紋狀體、皮層注射內(nèi)皮素-1制作大鼠腦缺血模型。大鼠隨機(jī)分為假手術(shù)組、模型組和dl-3-正丁基苯酞組。dl-3-正丁基苯酞組每日ig 70 mg/kg,起始于缺血后1周,持續(xù)給藥2周。采用平衡木實驗、粘性標(biāo)簽試驗和圓筒試驗評價神經(jīng)功能恢復(fù)情況,應(yīng)用免疫熒光技術(shù)觀察海馬齒狀回新生神經(jīng)細(xì)胞的情況。結(jié)果 dl-3-正丁基苯酞對腦缺血大鼠運功功能的恢復(fù)有明顯的改善,明顯促進(jìn)海馬齒狀回的新生神經(jīng)細(xì)胞增多,增加雙皮質(zhì)素(DCX)陽性細(xì)胞總樹突長度。結(jié)論 dl-3-正丁基苯酞不僅能夠?qū)δX缺血后大鼠的運動功能有明顯的改善,而且促進(jìn)腦缺血后大鼠的神經(jīng)再生,對腦缺血所致的神經(jīng)細(xì)胞損壞的治療提供參考。

Objective To explore the improvement of dl-3-n-butyphthalide on behavioral in rats with cerebral ischemia and its mechanism. Methods Cerebral ischemia model in rats was made by intracerebral injection of endothelin-1. Rats were divided into Sham group, model group, and dl-3-n-butyphthalide group. Rats in dl-3-n-butyphthalide group were ig administered with dl-3-n-butyphthalide 70 mg/kg. It began at 1 week after ischemia, and continued to be administered for 2 weeks. Tapered/ledged beam-walking test, cylinder test, and sticky label test were used to evaluate the functional recovery. Dentate gyrus regenerative nerve cells were observed by immunohistochemistry. Results dl-3-n-Butyphthalide can obviously improve the function recovery of cerebral ischemia in rats, significantly increased the number of new neurons in the dentate gyrus of hippocampus, and increase the total dendrite length of DCX positive cells. Conclusion dl-3-n-Butyphthalide can not only improve the motor function of rats after cerebral ischemia, but also promote the regeneration of nerve in rats after cerebral ischemia, which may provide reference for the treatment of nerve cell damage caused by cerebral ischemia.
