[摘要]
目的 研究芹菜素與γ-環(huán)糊精(γ-CD)包合物的制備、表征、包合機制及其抗氧化活性。方法 采用加熱回流共沉淀法制備芹菜素γ-環(huán)糊精包合物,紅外光譜法(IR)對形成的包合物進(jìn)行表征,熒光光譜法測定包合物的包合常數(shù)(K)和包合比(n);熒光光譜法和核磁共振法(NMR)研究芹菜素與γ-CD在溶液中形成的包合物;雙倒數(shù)曲線法研究包合前后對自由基1,1-二苯基-2-三硝基苯肼(DPPH·)的清除率。結(jié)果 在不同的pH值溶液中,γ-CD對芹菜素具有不同的包合能力,γ-CD最適合在中性介質(zhì)中與芹菜素形成包合物,在實驗濃度范圍內(nèi)按1:1形成包合物,包合比為1 236,包合物對自由基DPPH·的清除活性更強;NMR確定包合物的結(jié)構(gòu)。結(jié)論 在實驗條件下,芹菜素分子從γ-CD大口端進(jìn)入到空腔內(nèi)形成穩(wěn)定的包合物,且抗氧化活性增加。

[Abstract]
Objective To study the preparation, structure, inclusion mechanism and antioxidant activity of apigenin γ-cyclodextrin (γ-CD) inclusion complex. Methods Apigenin γ-cyclodextrin inclusion complex was prepared by heating reflux coprecipitation method. The inclusion complex was characterized by infrared spectroscopy (IR). The inclusion constant (K) and inclusion ratio (n) of the inclusion complex were determined. The inclusion complex of apigenin and γ-CD in solution was studied by fluorescence spectroscopy and nuclear magnetic resonance (NMR). The scavenging rate of 1,1-diphenyl-2-trinitrophenylhydrazine (DPPH·) before and after inclusion was studied by double reciprocal curve method. Results In different pH solutions, γ-CD has different inclusion ability to apigenin. γ-CD was most suitable to form inclusion complex with apigenin in neutral medium. In the range of experimental concentration, apigenin and γ-CD form inclusion complex was at 1:1, the inclusion ratio was 1 236, and apigenin/γ-CD inclusion complex was more stronger in scavenges free radical DPPH· than free apigenin. The structure of the inclusion complex was confirmed by NMR. Conclusion Under the experimental conditions, apigenin molecule is encapsulated into the cavity from the large mouth of γ-CD, forming a stable inclusion complex, and the antioxidant activity is increased.
