[摘要]
目的 探討益氣活血通絡(luò)方對蛛網(wǎng)膜下腔出血大鼠TGF-β/ERK信號通路的影響及神經(jīng)保護(hù)作用。方法 SD大鼠隨機(jī)分為假手術(shù)組,模型組,益氣活血通絡(luò)方5、10、15 g/kg組和尼莫地平組,除假手術(shù)組外,其余各組建立蛛網(wǎng)膜下腔出血模型大鼠,分組處理后,各組大鼠進(jìn)行神經(jīng)功能缺損評分;以伊文思藍(lán)染料外滲實(shí)驗(yàn)檢測大鼠血腦屏障通透性;以TUNEL染色檢測大鼠腦皮質(zhì)神經(jīng)細(xì)胞凋亡情況;以酶聯(lián)免疫吸附法(ELISA)檢測大鼠血清腫瘤壞死因子-α(TNF-α)、白細(xì)胞介素-6(IL-6)水平;以蛋白免疫印跡法檢測大鼠腦皮質(zhì)組織TGF-β/ERK通路相關(guān)蛋白TGF-β1、p-ERK/ERK表達(dá)情況。結(jié)果 與假手術(shù)組相比,模型組大鼠神經(jīng)功能缺損評分、伊文思藍(lán)滲出量、TUNEL陽性細(xì)胞比例、血清IL-6、TNF-α水平、TGF-β1表達(dá)、p-ERK/ERK明顯升高(P<0.05)。與模型組相比,益氣活血通絡(luò)方5、10、15 g/kg組和尼莫地平組大鼠神經(jīng)功能缺損評分、伊文思藍(lán)滲出量、TUNEL陽性細(xì)胞比例、血清IL-6、TNF-α水平、TGF-β1表達(dá)和p-ERK/ERK水平明顯降低(P<0.05),且益氣活血通絡(luò)方各組呈劑量相關(guān)性,益氣活血通絡(luò)方15 g/kg組與尼莫地平組相比,各指標(biāo)比較差異無統(tǒng)計(jì)學(xué)意義。結(jié)論 益氣活血通絡(luò)方可下調(diào)TGF-β/ERK信號,保護(hù)蛛網(wǎng)膜下腔出血大鼠神經(jīng)功能。

[Abstract]
Objective To investigate the neuroprotective effect of Yiqi Huoxue Tongluo Prescription on TGF-β/ERK signal pathway in rats with subarachnoid hemorrhage. Methods SD rats were randomly divided into sham operation group, model group, Yiqi Huoxue Tongluo Prescription Yiqi Huoxue Tongluo Prescription 5, 10, and 15 g/kg groups, and nimodipine group. In addition to the sham operation group, the other rats were established to establish subarachnoid hemorrhage model, after group treatment, the rats in each group were assessed for neurological deficit. Evans blue extravasation test was used to detect the permeability of BBB. TUNEL staining was used to detect the apoptosis of neurons. The levels of serum TNF-α and IL-6 were measured by enzyme-linked immunosorbent assay (ELISA), and the expressions of TGF-β1 and p-ERK/ERK in the cortex of rats were detected by Western blotting. Results Compared with the sham group, the neurological deficit score, seepage amount of Evans blue, proportion of TUNEL positive cell, levels of serum IL-6 and TNF-α, expression of TGF-β1, and p-ERK/ERK of rats in the model group were significantly increased (P<0.05). Compared with the model group, the neurological deficit score, seepage amount of Evans blue, proportion of TUNEL positive cell, levels of serum IL-6 and TNF-α, expression of TGF-β1 and p-ERK/ERK of rats in the Yiqi Huoxue Tongluo Prescription 5, 10, and 15 g/kg groups and nimodipine group were decreased (P<0.05), and Yiqi Huoxue Tongluo Prescription groups were dose-dependent, in addition, there was no significant difference in the indexes between Yiqi Huoxue Tongluo Prescription 15 g/kg group and nimodipine group. Conclusion Yiqi Huoxue Tongluo Prescription can down regulate TGF-β/ERK signal and protect the nerve function of rats with subarachnoid hemorrhage.
