[摘要]
D-氨基葡萄糖是人體內(nèi)天然存在的氨基己糖,也存在于甲殼類動(dòng)物的外骨骼中。D-氨基葡萄糖是糖蛋白和蛋白聚糖的重要組成部分,具有抗炎、抗腫瘤、抗氧化、抗纖維化和免疫調(diào)節(jié)等藥理作用,臨床上常用于緩解骨關(guān)節(jié)炎癥狀。目前已有大量文獻(xiàn)報(bào)道有關(guān)D-氨基葡萄糖抗腫瘤作用的研究成果,其抗腫瘤的作用機(jī)制主要包括抑制癌細(xì)胞增殖、誘導(dǎo)癌細(xì)胞凋亡和細(xì)胞周期阻滯、緩解炎癥反應(yīng)、誘導(dǎo)癌細(xì)胞自噬死亡、逆轉(zhuǎn)腫瘤耐藥性、抑制腫瘤血管生成、抑制基質(zhì)金屬蛋白酶的表達(dá)、調(diào)控轉(zhuǎn)錄因子以及免疫調(diào)節(jié)作用等。通過查閱國內(nèi)外相關(guān)文獻(xiàn)報(bào)道,綜述了D-氨基葡萄糖抗腫瘤作用機(jī)制的研究進(jìn)展,以期為D-氨基葡萄糖的深入研究提供參考。

D-glucosamine is a hexosamine that is produced naturally in human body, which is also found in the exoskeleton of crustaceans. It is an important component of glycoproteins and proteoglycans. D-glucosamine has many pharmacological properties, including anti-inflammatory, antitumor, antioxidant, anti-fibrosis and immune regulation. It is often used to relieve osteoarthritis in clinical. At present, numerous literatures have reported the research results of the antitumor activity of D-glucosamine, and the molecular mechanisms of which mainly include the suppression of tumor cell proliferation, promotion of apoptosis and arrest of the cell cycle, the suppression of inflammation, induction of autophagic cell death, reversal of drug-resistance, anti-angiogenic activity, inhibitory effect on matrix metalloproteinases expression, regulation of transcription factors, and immunomodulatory effects. This review summarizes antitumor molecular mechanisms of D-glucosamine in details, in order to provide the scientific basis for the further studies of D-glucosamine.
