炎癥反應(yīng)是機(jī)體常見的生理、病理活動(dòng),炎癥小體在該反應(yīng)中發(fā)揮重要調(diào)控作用。核苷酸結(jié)合寡聚化結(jié)構(gòu)域樣受體蛋白3(NOD-like receptor protein 3,NLRP3)是炎癥小體中關(guān)鍵的調(diào)控蛋白之一。研究發(fā)現(xiàn)激活NLRP3的刺激因子及其相關(guān)分子調(diào)控信號(hào)通路與多種疾病的發(fā)生、發(fā)展密切相關(guān),并越來越受到廣泛關(guān)注,是臨床藥物研究開發(fā)的前沿?zé)狳c(diǎn)方向??偨Y(jié)NLRP3炎癥小體的生物結(jié)構(gòu)、調(diào)控功能、信號(hào)通路和作用機(jī)制等研究進(jìn)展,并梳理與其相關(guān)靶點(diǎn)抑制劑的研究結(jié)果,對開展炎癥相關(guān)疾病的新靶標(biāo)藥物發(fā)現(xiàn)及其臨床應(yīng)用研究具有重要的參考價(jià)值。

[Abstract]
The inflammatory response is a common physiological and pathological process of the body, which is generally induced by the activation of various inflammatory factors. NLRP3 is a key regulatory protein. It has been found that there are a variety of stimulating factors and molecular regulatory signal pathways involved in NLRP3 activation closely related to the occurrence and development of a variety of diseases, and have attracted more and more attention. It is one of the frontier hot topics in clinical drug research and development. This paper summarizes the research progress in biological structure, regulatory pathway and inhibitors development of NLRP3 inflammasome, which is helpful for the target seeking and clinical drug development.
