[摘要]
目的 研究乳香及其炮制品對葡聚糖硫酸鈉(DSS)誘導的小鼠炎癥性腸病的保護作用。方法 采用DSS水溶液代替正常飲水建立炎癥性腸病小鼠模型,將實驗小鼠分為對照組、模型組、柳氮磺胺吡啶(300 mg/kg)組、生乳香(0.76 g/kg)組、醋乳香(0.76 g/kg)組、生乳香糞清液(0.2 mL)組、醋乳香糞清液(0.2 mL)組,分別進行給藥治療。觀察和監(jiān)測小鼠體質(zhì)量、糞便性狀、便血情況,進行疾病活動指數(shù)(DAI)評分;記錄結腸長度;運用蘇木精–伊紅染色法(HE)觀察炎癥性腸病小鼠腸道組織的病理變化;酶聯(lián)免疫吸附試驗(ELISA)檢測炎癥性腸病小鼠血清中白細胞介素(IL)-4、IL-10、IL-6、IL-1β的水平。采用蛋白質(zhì)免疫印跡法(Western blotting)測定炎癥性腸病小鼠腸道組織中腸黏膜機械屏障蛋白的表達情況。結果 生乳香、醋乳香、生乳香糞清液組和醋乳香糞清液組均可改善DSS造成的小鼠體質(zhì)量下降、糞便性狀評分和便血情況,顯著降低DAI評分;改善炎癥性腸病小鼠腸道組織的病理狀態(tài);調(diào)節(jié)血清中的炎癥因子水平;顯著升高炎癥性腸病小鼠腸道組織中咬合蛋白(occludin)、緊密連接蛋白1(claudin-1)、閉合小環(huán)蛋白1(ZO-1)蛋白表達水平。結論 乳香及其炮制品可改善炎癥性腸病小鼠炎癥反應,對腸黏膜屏障具有一定的保護作用,且糞菌移植效果更佳。

Objective To investigate the protective effects of Olibanum and its processed products on inflammatory bowel disease induced by dextran sulfate sodium (DSS) in mice.Methods DSS solution was used instead of normal drinking to establish inflammatory bowel disease mice model, experimental mice were divided into control group, model group, sulfasalazine (300 mg/kg) group, raw Olibanum (0.76 g/kg) group, vinegar Olibanum (0.76 g/kg) group, raw Olibanum purified feces (0.2 mL) group, and vinegar Olibanum purified feces (0.2 mL) group. The body weight, fecal traits, and hematochezia of mice were observed and monitored, and disease activity index (DAI) was scored. Colon length was recorded. HE staining was used to observe the pathological changes of intestinal tissues in mice with inflammatory bowel disease. The levels of interleukin-4, IL-10, IL-6, and IL-1β in serum of mice with inflammatory bowel disease were determined by ELISA. Western blotting was used to detect the protein expression in intestinal tissues of inflammatory bowel disease mice. Results Raw Olibanum, vinegar Olibanum, raw Olibanum purified feces, and vinegar Olibanum purified feces group could improve the decrease of body weight, fecal traits score, and blood in stool caused by DSS, and significantly reduce DAI score. To improve the pathological state of intestinal tissue in mice with inflammatory bowel disease. Regulating the level of inflammatory factors in serum. The expression levels of occludin, claudin-1, and ZO-1 in intestinal tissues of inflammatory bowel disease mice were significantly increased. Conclusion Olibanum and its processed products can improve the inflammatory response of inflammatory bowel disease mice and have certain protective effect on intestinal mucosal barrier.
