[摘要]
目的 探討以多酚類天然產(chǎn)物為組方的傳世逆甲片對(duì)腫瘤早篩基因甲基化的干預(yù)調(diào)控作用特點(diǎn)。方法 采用甲基化特異性定量聚合酶鏈?zhǔn)椒磻?yīng)(qMSP),針對(duì)從文獻(xiàn)中篩選得到的腫瘤早篩基因波形蛋白(VIM)、前列腺素E受體4(PTGER4)、矮小同源盒基因(SHOX2)、骨形態(tài)發(fā)生蛋白3(BMP3)、胞裂蛋白9(SEPT9)基因,設(shè)計(jì)甲基化特異性引物。選取肺癌NCI-H446細(xì)胞作為研究對(duì)象,分別經(jīng)20μg/mL的姜黃素、茶多酚、茶色素、傳世逆甲片和2μmol/L 5-氮雜胞苷作用后,提取全基因組DNA,得到其基因的啟動(dòng)子區(qū)的甲基化相對(duì)值。結(jié)果 多酚類天然產(chǎn)物姜黃素和5-氮雜胞苷對(duì)PTGER4、SHOX2、SEPT9的啟動(dòng)子甲基化具有抑制作用,而茶色素對(duì)PTGER4、BMP3、SHOX2、SEPT9基因啟動(dòng)子的甲基化具有促進(jìn)作用,傳世逆甲片對(duì)SHOX2和SEPT9甲基化抑制作用最強(qiáng)。結(jié)論 傳世逆甲片及其含有的多酚類物質(zhì)能夠抑制腫瘤早期篩查基因甲基化水平,干預(yù)調(diào)控腫瘤早篩基因甲基化作用。

[Abstract]
Objective To investigate the inhibitory effect of Nijia (TRANS-ME) Recipe, which is grouped with polyphenolic natural products, on methylation of early tumor screening genes, and to provide an experimental basis for the study of natural polyphenols in tumor prevention. Methods Quantitative methylation-specific polymerase chain reaction (qMSP) was used. Methylation-specific primers were designed for the early tumor screening genes VIM, PTGER4, SHOX2, BMP3, and SEPT9 genes obtained from literature screening. Lung cancer NCI-H446 cells were selected for the study, after being treated with 20 μg/mL curcumin, tea polyphenol, tea pigment, Nijia (TRANS-ME) Recipe, and 2 μmol/L 5-azocytoside, whole genomic DNA was extracted, and then the relative methylation values of the promoter regions of their genes were obtained. Results Curcumin and 5-azocytoside inhibit the promoter methylation of PTGER4, SHOX2 and SEPT9, while tea pigment promotes the promoter methylation of PTGER4, BMP3, SHOX2, and SEPT9. The methylation inhibition of Nijia (TRANS-ME) Recipe of SHOX2 and SEPT9 was strongest. Conclusion Nijia (TRANS-ME) Recipe and the polyphenols contained in them can inhibit the methylation level of tumor early screening genes and regulate the methylation of tumor early screening genes.
