[摘要]
目的 探討芩百清肺濃縮丸對(duì)肺炎支原體肺炎小鼠肺上皮間質(zhì)轉(zhuǎn)化的影響。方法 將Balb/c小鼠隨機(jī)分為對(duì)照組、模型組、阿奇霉素(0.03 g/kg)組及芩百清肺濃縮丸高、中、低劑量(4.6、2.3、1.15 g/kg)組,每組10只。除對(duì)照組外,其余各組小鼠進(jìn)行支原體滴鼻,每只滴入20 μL 1×107 CCU肺炎支原體菌液,建立肺炎支原體感染模型。支原體肺炎模型建立成功后各組分別ig給予小鼠相應(yīng)藥物,連續(xù)ig 14 d。實(shí)驗(yàn)結(jié)束后取出小鼠肺組織,顯微鏡下觀察各組小鼠肺組織蘇木精–伊紅(HE)染色、Masson染色以及免疫組化的肺組織變化。RT-PCR法檢測(cè)各組小鼠肺組織E-鈣黏蛋白(E-cadherin)、轉(zhuǎn)化生長(zhǎng)因子-β1(TGF-β1)和波形蛋白(Vimentin)mRNA表達(dá)量。結(jié)果 與模型組相比,芩百清肺濃縮丸組小鼠肺組織結(jié)構(gòu)明顯得到改善,結(jié)構(gòu)清晰,組織形態(tài)規(guī)整。與模型組比較,芩百清肺濃縮丸組小鼠肺組織E-cadherin mRNA及蛋白表達(dá)增高;TGF-β1、Vimentin mRNA及蛋白表達(dá)顯著降低(P<0.05)。結(jié)論 芩百清肺濃縮丸能夠降低肺組織中TGF-β1、Vimentin的表達(dá),增加E-cadherin的表達(dá),從而抑制上皮間質(zhì)轉(zhuǎn)化。

[Abstract]
Objective To investigate the effect of Qinbai Qingfei Concentrated Pills on pulmonary epithelial interstitial transformation in mice with mycoplasma pneumoniae pneumonia. Methods Balb/c mice were randomly divided into control group, model group, azithromycin (0.03 g/kg) group and Qinbai Qingfei Concentrated Pills high, medium, and low-dose (4.6, 2.3, 1.15 g/kg) groups, with 10 mice in each group. Except the control group, mice in other groups received mycoplasma nasal drops, each of which was injected with 20 μL 1× 107 CCU mycoplasma pneumoniae solution to establish mycoplasma pneumoniae infection model. After the successful establishment of mycoplasma pneumonia model, mice in each group were given corresponding drugs intragastrically and intragastrically for 14 d. At the end of the experiment, the lung tissues of mice were taken out, and the changes of the lung tissues with hematoxylin-eosin (HE) staining and Masson staining were observed under the microscope. After the experiment, the lung tissues of mice were taken out, and the changes of hematoxylin-eosin (HE) staining, Masson staining, and immunohistochemical lung tissue were observed under the microscope. The mRNA expressions of E-cadherin, TGF-β1, and Vimentin in lung tissues of mice in each group were detected by Real-time PCR. Results Compared with model group, the lung tissue structure of mice in Qinbai Qingfei Concentrated Pills group was obviously improved, with clear structure and regular morphology. Compared with model group, mRNA and protein expression of E-cadherin in lung tissue of mice in Qinbai Qingfei Concentrated Pills group were increased. The mRNA and protein expressions of TGF-β1 and Vimentin were significantly decreased (P < 0.05). Qinbaiqingfei concentrated pills can decrease the expression of TGF-β1 and Vimentin, and increase the expression of E-cadherin in lung tissue, so as to inhibit the epithelial mesenchymal transformation.Conclusion Qinbai Qingfei Concentrated Pills can decrease the expression of TGF-β1 and Vimentin, and increase the expression of E-cadherin in lung tissue, so as to inhibit the epithelial mesenchymal transformation.
