[摘要]
目的 探討五味子提取物對小鼠銀屑病樣皮損的抑制作用及機制。方法 使用咪喹莫特乳劑建立小鼠銀屑病背部皮損模型,將小鼠分為對照組、模型組、他克莫司5 mg/kg組以及五味子提取物0.05、0.25 mg/kg組,并于造模后在小鼠背部皮膚涂抹相應藥物。取小鼠背部皮膚組織,分析皮損癥狀,對其進行蘇木精–伊紅(HE)染色,并觀察測定表皮厚度。免疫組織化學測定淋巴細胞表面分子(CD-8)、環(huán)氧化酶-2(COX-2)、細胞外調(diào)節(jié)蛋白激酶(ERK)、磷酸化細胞外調(diào)節(jié)蛋白激酶(p-ERK)的表達情況。建立3D皮膚模型,使用雙氧水造模后,噻唑藍(MTT)檢測五味子提取物抗氧化修復活性、酶聯(lián)免疫吸附法檢測皮膚模型組織中超氧化物歧化酶(SOD)和過氧化氫酶(CAT)的酶活力和炎癥因子白細胞介素-6(IL-6)的含量。結果 五味子提取物可以緩解由咪喹莫特乳劑誘導的小鼠銀屑病皮膚損傷、紅斑、炎癥,改善其皮損處組織細胞情況,降低銀屑病模型小鼠表皮厚度(P<0.05);五味子提取物可以下調(diào)銀屑病模型小鼠皮膚組織中COX-2、ERK、p-ERK、CD-8的表達(P<0.05、0.01);3D皮膚模型檢測結果顯示,五味子提取物可以減緩雙氧水對3D皮膚細胞的氧化損傷作用,降低皮膚組織細胞中IL-6的表達,同時可以提高SOD和CAT酶的活性(P<0.05、0.01)。結論 五味子提取物對小鼠銀屑病樣皮損具有一定抑制作用,可能與通過免疫調(diào)節(jié)來抑制CD-8、COX-2、ERK、p-ERK的表達和緩解氧化應激損傷有關。

[Abstract]
Objective To investigate the inhibitory effect and mechanism of Schisandra Chinensis Fructus extract on psoriasis-like skin lesions in mice. Methods Mice were divided into control group, model group, tacrolimus 5 mg/kg group, and Schisandra Chinensis Fructus extract 0.05 and 0.25 mg/kg groups. After modeling, the mice were smeared with the corresponding drug on the skin of the back of the mice. The skin tissue of the back of mice was collected, the symptoms of skin lesions were analyzed, and the skin thickness was observed and measured by hematoxylin-eosin (HE) staining. The expressions of CD-8, COX-2, ERK, and p-ERK were determined by immunohistochemistry. The 3D skin model was established, and the antioxidant repair activity of Schisandra Chinensis Fructus extract was detected by thiazole blue (MTT), and the enzyme activity of SOD, CAT, and the content of inflammatory factor IL-6 in the skin model tissues were detected by enzymelinked immunosorbent assay (ELISA). Results Schisandra Chinensis Fructus extract can relieve the skin injury, erythema and inflammation induced by imiquimod emulsion, improve the tissue and cell condition of the lesions, and reduce the epidermal thickness of psoriasis model mice (P < 0.05). The extract of Schisandra Chinensis Fructus extract could down-regulate the expressions of COX-2, ERK, p-ERK, and CD-8 in the skin tissue of psoriasis model mice (P < 0.05, 0.01). 3D skin model test results showed that Schisandra Chinensis Fructus extract can slow down the oxidative damage of hydrogen peroxide on 3D skin cells, reduce the expression of IL-6 in skin tissue cells, and increase the activities of SOD and CAT enzymes (P < 0.05, 0.01). Conclusion The extract of Schisandra Chinensis Fructus extract showed certain inhibitory effect on psoriasis lesions in mice, which may be related to the inhibition of CD-8, COX-2, ERK, and p-ERK expression and the relief of oxidative stress injury through immune regulation.
