[摘要]
目的 采用高效液相色譜-四級(jí)桿/靜電場(chǎng)軌道阱高分辨質(zhì)譜(HPLC-Q-Exactive-Orbitrap-MS)法鑒定森登-4味涂劑中化學(xué)成分。方法 色譜條件:Shim-pack GIST-HP C18色譜柱(150 mm×4.6 mm,5 μm),流動(dòng)相為乙腈-0.1%甲酸水溶液,梯度洗脫,體積流量0.35 mL/min,柱溫為40 ℃,進(jìn)樣量為5 μL。質(zhì)譜條件:檢測(cè)方式Full MS/dd-MS2,掃描范圍m/z 110~1 200。電噴霧離子源(ESI),正、負(fù)離子模式掃描,噴霧電壓4.20 kV(+)、3.80 kV(-),碰撞能量(CE)30 eV。通過(guò)全方化學(xué)成分信息庫(kù),結(jié)合多級(jí)質(zhì)譜碎片信息、對(duì)照品保留時(shí)間,鑒定森登-4味涂劑中化學(xué)成分。結(jié)果 在森登-4味涂劑中共鑒定出60個(gè)成分,包括16個(gè)黃酮類(lèi)成分、13個(gè)有機(jī)酸類(lèi)成分、11個(gè)環(huán)烯醚萜類(lèi)成分、10個(gè)鞣質(zhì)類(lèi)成分、4個(gè)氨基酸類(lèi)成分、4個(gè)脂肪酸類(lèi)成分、2個(gè)三萜類(lèi)成分,并考察了山梔苷、槲皮素、阿魏酸、克里拉京、齊墩果酸、脯氨酸、亞油酸的MS2圖和可能的裂解途徑。結(jié)論 對(duì)森登-4味涂劑中化學(xué)成分進(jìn)行了快速、準(zhǔn)確的鑒定,為其質(zhì)量標(biāo)準(zhǔn)建立、藥效物質(zhì)挖掘和作用機(jī)制研究提供了實(shí)驗(yàn)基礎(chǔ)。

[Abstract]
Objective To identify the chemical components in Sendeng-4 Pigmentum by HPLC-Q-Exactive-Orbitrap-MS. Methods Chromatographic conditions: Shim-pack GIST-HP C18 column (150 mm × 4.6 mm, 5 μm) was employed with the mobile phase of acetonitrile - 0.1% formic acid solution for gradient elution. The flow rate was 0.35 mL/min, the column temperature was set at 40 ℃, and the volume of sample injection was 5 μL. Mass spectrometry conditions: Detection method: Full MS/dd MS2, scanning range m/z 110 — 1 200. Electrospray ionization (ESI) was applied for scanning positive and negative ion mode scanning with spray voltage 4.20 kV (+) and 3.80 kV (-), and collision energy was 30 eV. Comprehensive chemical composition information database, combined with multi-level mass spectrometry fragment information and reference substance retention time were used to identify the chemical components in Sendeng-4 Pigmentum. Results A total of 60 components were identified in Sendeng-4 Pigmentum, including 16 flavonoids, 13 organic acids, 11 iridoids, 10 tannins, 4 amino acids, 4 fatty acids, and 2 triterpenes. MS2 and possible fragmentation pathway of shanzhiside, quercetin, ferulic acid, corilagin, oleanolic acid, proline, and linoleic acid were investigated. Conclusion Chemical components in Sendeng-4 Pigmentum were quickly and accurately identified, providing an experimental basis for the establishment of its quality standards, the exploration of pharmacological substances, and its mechanism of action.
