[摘要]
目的 探討芩百清肺濃縮丸對(duì)肺炎支原體CARDs TX毒素蛋白表達(dá)的抑制作用。方法 將Balb/c小鼠隨機(jī)分為對(duì)照組、模型組、阿奇霉素組及芩百清肺濃縮丸4.6、2.3、1.15 g/kg組,每組9只。除對(duì)照組外,其余各組小鼠進(jìn)行支原體滴鼻,每只滴入20 μL 1×106 CCU肺炎支原體菌液,建立肺炎支原體感染模型。支原體肺炎模型建立成功后各組分別ig小鼠相應(yīng)藥物,連續(xù)ig 7 d,末次給藥結(jié)束后進(jìn)行引咳實(shí)驗(yàn),并處死小鼠,取血液與肺組織。顯微鏡下觀(guān)察各組小鼠肺組織蘇木精–伊紅(HE)染色、Masson染色、免疫組化后的肺組織變化;RT-PCR法檢測(cè)各組小鼠肺組織MPN372、BDKRB2表達(dá)量。結(jié)果 與模型組相比,芩百清肺濃縮丸可顯著降低肺炎支原體小鼠咳嗽次數(shù);芩百清肺濃縮丸組小鼠肺組織結(jié)構(gòu)明顯得到改善,結(jié)構(gòu)清晰,組織形態(tài)規(guī)整;芩百清肺濃縮丸組小鼠肺組織纖維化得到緩解;芩百清肺濃縮丸組小鼠肺組織MPN372、BDKRB2 mRNA表達(dá)顯著降低(P<0.05)。結(jié)論 芩百清肺濃縮丸通過(guò)降低肺組織中CARDs TX、BDKRB2的表達(dá),減緩咳嗽癥狀,治療肺炎支原體肺炎。

[Abstract]
Objective To investigate the inhibitory effect of Qinbai Qingfei Concentrated Pills on the expression of CARDs TX toxin protein of mycoplasma pneumoniae. Methods Balb/c mice were randomly divided into control group, model group, azithromycin group, and Qinbai Qingfei Concentrated Pills (4.6, 2.3 and 1.15 g/kg) groups, with 9 mice in each group. Except the control group, mice in other groups received mycoplasma nasal drops, each of which was infused with 20 μL 1×106 CCU mycoplasma pneumoniae solution, and the mycoplasma pneumoniae infection model was established. After the successful establishment of mycoplasma pneumonia model, mice in each group were ig treated with the corresponding drug for 7 d. After the last administration, the cough inducing experiment was carried out, and the mice were killed, and blood and lung tissues were collected. The changes of lung tissue after HE staining, Masson staining, and immunohistochemistry were observed under microscope. The expression levels of MPN372 and BDKRB2 in lung tissues of each group were detected by RT-PCR. Results Compared with the model group, Qinbai Qingfei Concentrated Pills group can significantly reduce the cough times of mycoplasma pneumoniae mice. In Qinbai Qingfei Concentrated Pills group, the lung tissue structure was obviously improved, the structure was clear and the tissue morphology was regular. The pulmonary fibrosis of mice in Qinbai Qingfei Concentrated Pills group was alleviated. The mRNA expressions of MPN372 and BDKRB2 in lung tissue of Qinbai Qingfei Concentrated Pills group were significantly decreased (P < 0.05). Conclusion Qinbai Qingfei Concentrated Pills can reduce the expression of CARDs TX and BDKRB2 in lung tissue to relieve cough symptoms and treat mycoplasma pneumoniae pneumonia.
