[摘要]
目的 建立高效液相色譜-一測多評法測定連花清瘟膠囊中綠原酸、甘草苷、木犀草苷、連翹酯苷A、連翹苷、大黃酸的方法。方法 使用Inertsil ODS-3色譜柱(250 mm×4.6 mm,5 μm);流動相:乙腈-0.4%磷酸水,梯度洗脫;體積流量:1.0 mL/min;柱溫:25 ℃;檢測波長:280 nm。以連翹苷為內(nèi)參物,建立其他5種成分的相對校正因子,一測多評法計(jì)算各成分。結(jié)果 綠原酸、甘草苷、木犀草苷、連翹酯苷A、連翹苷、大黃酸分別在0.243~4.861、0.112~2.248、0.087~1.732、0.247~4.949、0.370~7.402、0.095~0.952μg/mL線性關(guān)系良好(r≥0.999 5),平均回收率分別為99.40%、101.0%、104.5%、98.17%、99.70%、99.58%,RSD值分別為1.12%、2.53%、3.15%、2.94%、1.41%、2.84%(n=6)。結(jié)論 該法回收率高、重復(fù)性良好,為連花清瘟膠囊建立更全面的質(zhì)量控制提供了新方法。

[Abstract]
Objective To establish an HPLC-quantitative analysis of multi-components by single marker (QAMS) to determine contents of chlorogenic acid, glycyrrhizin, luteolin, forsythiaside A, forsythin, and rhein in Lianhua Qingwen Capsules. Methods Analysis was performed on an Inertsil ODS-3 column (250 mm × 4.6 mm, 5 μm). The mobile phase consisted of acetonitrile-0.4% phosphoric acid aqueous in gradient mode. The flow rate was 1.0 mL/min, the column temperature was maintained at 25℃, and the detection wavelength was set at 280 nm. Using forsythin as the internal reference substance, the relative correction factor of the other five components was established, and the contents of each component were calculated by QAMS method. Results Chlorogenic acid, glycyrrhizin, luteolin, forsythiaside A, forsythin, and rhein all showed good linear relationship within the ranges of 0.243-4.861, 0.112-2.248, 0.087-1.732, 0.247-4.949, 0.370-7.402, and 0.095-0.952 μg/mL ( r ≥ 0.999 5), and their average recovery rates were 99.40%, 101.0%, 104.5%, 98.17%, 99.70%, and 99.58%, with the RSD of 1.12%, 2.53%, 3.15%, 2.94%, 1.41%, and 2.84%(n=6)。Conclusion This method has a high recovery rate and good repeatability, providing a new method for establishing a more comprehensive quality control of Lianhua Qingwen Capsules.
