18柱(150 mm×4.6 mm,5 μm),流動相為0.05%磷酸-乙腈(65∶35),檢測波長254 nm,常溫,進樣量50 μL。以靈芝多糖肽為對照品,采用Folin-酚試劑法,通過紫外-可見吸收光譜法檢測菌草靈芝和仿野生栽培靈芝不同收獲期提取物的多糖肽。結(jié)果 靈芝酸A、B線性范圍分別為14.8~371.0和16.4~409.0 μg/mL,平均回收率為95.0%、100.5%,RSD分別為1.18%、3.37%(n=6)。靈芝多糖肽在100~600 μg/mL與吸光度值呈線性關(guān)系,r=0.999 5,回收率95%~100%。結(jié)論 該方法簡便、快速,可用于靈芝及其產(chǎn)品中多糖肽和靈芝酸A、B的質(zhì)量控制。菌草靈芝沸水提取能將多糖肽和靈芝酸分離,既簡化了靈芝水、醇提取工藝,又提高經(jīng)濟效益。;Objective To study the changes of Ganoderma lucidum polysaccharide peptides (GLPP) and ganoderic acids A and B by different cultivation methods, which provides the basis for G. lucidum standardized cultivation and establishment of its product quality standard. Methods To determine the changes of ganoderic acids A and B between grass-cultivated G. Lucidum (GCGL) and wood logs cultivated G. lucidum (WLCGL) extracts by RP-HPLC method. Chromatography on XB-C18 column (150 mm × 4.6 mm, 5 μm) was used and the mobile phase was 0.05% phosphoric acid-acetonitrile (65:35) with detection wavelength of 254 nm at ambient temperatures and sample quantity of 50 μL. With GLPP as reference, the content of polysaccharide peptide from GCGL and WLCGL extracts was tested in UV-VIS by using the method of Folin-phenol reagent. Results Linear ranges of ganoderic acids A and B were 14.8—371.0 and 16.4—409.0 μg/mL, respectively. The average recovery rate of ganoderic acids A and B were 95.0% and 100.5%, and RSD were 1.18% and 3.37% (n = 6), respectively. In the range of 100—600 μg/mL, linear relationship was between concentrations and absorbance of GLPP, r = 0.999 5, with the recovery rate of 95%—100%. Conclusion The method is simple and convenient, which could be used for the quality control of GLPP and the changes of ganoderic acid A and B. It is found that GCGL could be extracted from boiling water, which could separate GLPP and ganoderic acids at the same time. It could not only simplify the alcohol and water extraction process of G. lucidum, but also improve the economic efficiency."/>
