目的 觀察大黃素對(duì)人胃癌MNK-45和MGC8-03細(xì)胞增殖的影響。方法 采用MTT法測(cè)定MNK-45細(xì)胞和MGC8-03細(xì)胞增殖抑制率；采用AO/EB雙染觀察細(xì)胞形態(tài)學(xué)變化；采用流式細(xì)胞儀測(cè)定細(xì)胞周期。結(jié)果 大黃素（5～80 μg/mL）對(duì)MNK-45和MGC8-03細(xì)胞增殖均具有顯著抑制作用。P＜0.01或0.05），且藥物劑量越大，作用時(shí)間越長(zhǎng)，其抑制作用越強(qiáng)（P＜0.01）；大黃素作用于兩種細(xì)胞后熒光顯微鏡下觀察均顯示細(xì)胞凋亡特征；大黃素使MNK-45和MGC8-03細(xì)胞增殖被阻滯于G₀/G₁期。結(jié)論 大黃素能抑制人胃癌MNK-45和MGC8-03細(xì)胞增殖，誘導(dǎo)其凋亡，影響其細(xì)胞周期時(shí)相的分布。

Objective To observe the effects of emodin on the proliferation of human gastric carcinoma cell lines MNK-45 and MGC8-03. Methods MTT assay was used to measure the inhibitory rate on the cell growth of MNK-45 and MGC8-03. The morphological changes were determined using AO/EB staining. The changes of cell cycle were detected by flow cytometry. Results Emodin (5―80 μg/mL) had the significant inhibition on cell proliferation of MNK-45 and MGC8-03 (P < 0.01 or 0.05). In addition, the higher the dosage was, the longer the effective time was, and the stronger the inhibiton was (P < 0.01); After emodin treatment, the two cell lines showed the features of apoptosis; Emodin could make the cell proliferation of MNK-45 and MGC8-03 stay in the G₀/G₁ phase. Conclusion Emodin could inhibit the cell proliferation of human gastric carcinoma MNK-45 and MGC-803, induce the apoptosis of MNK-45 and MGC-803, and affect the distribution of cell cycle phase.
