[摘要]
目的 研究益母草注射液浸膏及其提取部位對(duì)大鼠流產(chǎn)子宮的作用。方法 采用化學(xué)提取分離方法由益母草注射液中提取分離得到益母草生物堿部位(LH-1-B)、非生物堿部位(LH-1-A)。早孕大鼠ig米非司酮(8.3 mg/kg)和米索前列醇(100 μg/kg)造成不完全流產(chǎn),24 h后頸椎脫臼處死動(dòng)物,迅速剖取子宮,用BL-420E生物機(jī)能實(shí)驗(yàn)系統(tǒng)記錄大鼠離體子宮平滑肌的收縮活動(dòng);采用累積給藥方式,分別給予益母草注射液浸膏(37.5、37.5、150.0 μL)或LH-1-A、LH-1-B(75.0、75.0、 150.0 μL),每次給藥后記錄10 min收縮曲線,比較給藥前后收縮張力、頻率和活力的變化。結(jié)果 益母草注射液浸膏可使流產(chǎn)大鼠離體子宮的活力和收縮張力增強(qiáng)(P<0.01);LH-1-A使流產(chǎn)大鼠離體子宮收縮活力、張力及頻率降低(P<0.05、0.01);LH-1-B高劑量具有增加流產(chǎn)子宮收縮張力最小值作用(P<0.05)。結(jié)論 益母草注射液浸膏可顯著增強(qiáng)流產(chǎn)后大鼠子宮收縮活動(dòng),但其內(nèi)含有促進(jìn)流產(chǎn)子宮收縮和抑制流產(chǎn)子宮收縮的活性部位,其中水溶性生物堿部位是其縮宮的主要部位,作用機(jī)制有待進(jìn)一步研究。

[Abstract]
Objective To study the effects of the extract from Leonurus japonicus Injection (LJI) on uterus of rats after abortion. Methods The alkaloids fraction (LH-1-B) and non-alkaloid fraction (LH-1-A) were separated from LJI by chemical extraction. Pre pregnancy rats were ig administered with Mifepristone (8.3 mg/kg) and Misoprostol (100 μg/kg) to induce incomplete abortion model. After 24 h, the rats were executed with cervical dislocation and the uterus was taken. The contraction activity of uterine smooth muscle in rats was recorded by BL—420E Biological Function Experimental System. The doses of extracts were 37.5, 75, and 150 μL and the doses of LH-1-A and LH-1-B were 75, 150, and 300 μL by using cumulative mode of administration. The constriction curve for 10 min was recorded including the amplitude, frequency, and mobility in uterine smooth muscle movement track. Results The extract from LJI could enhance the activity of abortion uterus (P < 0.01); The LH-1-A extract could decrease the activity of abortion uterus (P < 0.05 or 0.01); The high-dose LH-1-B could increase the minimum of uterus contraction (P < 0.05). Conclusion The extract from LJI could significantly enhance the contraction of uterus in rats after abortion, and contains the components with the effects of promoting and inhibiting contraction of uterus. The alkaloid fraction is the main part with the promoting effect. Further investigations are needed to explore the action mechanism.
