[摘要]
目的 采用星點設(shè)計效應(yīng)面法優(yōu)化川芎中阿魏酸和藁本內(nèi)酯的提取工藝。方法 以川芎藥材中阿魏酸、藁本內(nèi)酯的質(zhì)量分?jǐn)?shù)為考察指標(biāo),考察乙醇體積分?jǐn)?shù)、提取溫度以及提取時間對提取工藝的影響,對結(jié)果進行二項式擬合,用效應(yīng)面法選擇較佳工藝條件,并進行預(yù)測分析。結(jié)果 確定較優(yōu)提取工藝為83 ℃下,14倍量73%乙醇提取2次,每次2 h,含量實際值與預(yù)測值偏差為?1.37%。結(jié)論 星點設(shè)計效應(yīng)面法優(yōu)化川芎提取工藝,方法簡便,結(jié)果可靠,預(yù)測性良好。

[Abstract]
Objective To optimize the extraction process of ferulaic acid and ligustilide in Ligusticum chuanxiong by central composite design and response surface method. Methods The effects of ethanol concentration, extracting temperature, and extracting time on the contents of ferulaic acid and ligustilide were investigated by using central composite design and response surface method. The data were imitated using multi-linear equation and second-order polynomial equation. Results The optimal conditions of extraction process were 14 fold solvent, 2 h for decoction, and extracting twice; The ethanol concentration was 73%, the extracting temperature was 83 ℃. The variance between observed and predicted values was ?1.37%. Conclusion The central composite design and response surface method is convenient, reliable, and highly predictive.
