[摘要]
目的 觀察如意珍寶丸對(duì)斑馬魚神經(jīng)損傷的保護(hù)與促再生作用。方法 用嗎替麥考酚酯誘導(dǎo)斑馬魚中樞神經(jīng)損傷模型;用乙醇誘導(dǎo)建立斑馬魚外周運(yùn)動(dòng)神經(jīng)損傷及軸索損傷模型;用溴化乙錠誘導(dǎo)建立斑馬魚髓鞘損傷模型。分別在熒光顯微鏡下觀察高、中、低質(zhì)量濃度(10.0、33.3和100.0 μg/mL)的如意珍寶丸對(duì)模型斑馬魚的中樞神經(jīng)、軸索熒光強(qiáng)度、外周運(yùn)動(dòng)神經(jīng)長(zhǎng)度以及髓鞘熒光強(qiáng)度變化的影響,利用NIDS-Element's軟件分別進(jìn)行圖像分析,計(jì)算如意珍寶丸對(duì)斑馬魚中樞神經(jīng)、軸索的保護(hù)率、對(duì)外周運(yùn)動(dòng)神經(jīng)及髓鞘再生的促進(jìn)率。結(jié)果 質(zhì)量濃度分別為10.0、33.3和100.0 μg/mL的如意珍寶丸對(duì)斑馬魚中樞神經(jīng)損傷保護(hù)率分別為-2%、24%和50%(P<0.001);軸索損傷的保護(hù)率分別為3%、29%和48%(P<0.05);外周神經(jīng)再生促進(jìn)率分別為44%(P < 0.05)、49%(P < 0.01)和93%(P < 0.001);髓鞘再生促進(jìn)率分別為36%(P < 0.01)、37%(P < 0.001)和41%(P < 0.001)。結(jié)論 如意珍寶丸對(duì)斑馬魚中樞神經(jīng)及軸索損傷具有保護(hù)作用,對(duì)斑馬魚外周運(yùn)動(dòng)神經(jīng)損傷及髓鞘損傷具有促再生作用。

[Abstract]
Objective To observe the protective and regeneration-promoting effects of Ruyi Zhenbao Pill (RZP) on nerve injury in zebrafish. Methods The zebrafish model of central nervous injury was induced by mycophenolate mofetil, the model of peripheral motor nerve and axonal injury was induced by ethanol, and the model of myelin damage was induced by ethidium bromide. The variations of central nerve, axon and myelin sheath fluorescence intensity, and peripheral motor nerve length in zebrafish, which exposed to the different concentration (10.0, 33.3, and 100.0 μg/mL) of RZP, were observed with fluorescence microscope. The effective protection rates of RZP on zebrafish central nerve and axone, and the regeneration-promoting effect on peripheral motor nerve and myelin sheath were analyzed and calculated with the image processing software NIDS-Element's. Results In 10.0, 33.3, and 100.0 μg/mL RZP groups, the zebrafish central nervous injury protective rates were 2%, 24%, and 50% (P<0.001), respectively, the peripheral nerve regeneration promoting rates were 44% (P < 0.05), 49% (P < 0.01), and 93% (P < 0.001), the axonal injury recovery rates were 3%, 29% and 48% (P<0.05), and the myelin sheath regeneration promoting rates were 36% (P < 0.01), 37% (P < 0.001), and 41% (P < 0.001), compared with model group. Conclusions RZP could not only protect the central nervous and axonal injury, but also promote the regeneration of peripheral nerve and myelin sheath in zebrafish.
