[摘要]
目的研究中成藥潤(rùn)肺止嗽丸對(duì)慢性阻塞性肺疾病(COPD)大鼠細(xì)胞凋亡的影響,并探討其作用機(jī)制。方法 32只雄性Wistar大鼠隨機(jī)分為對(duì)照組、模型組、潤(rùn)肺1月和3月組;使用煙熏聯(lián)合氣道滴入脂多糖法制備COPD大鼠模型;潤(rùn)肺1、3月組ig給予潤(rùn)肺止嗽丸0.9 g/kg,每天給藥1次,分別連續(xù)給藥1、3個(gè)月,對(duì)照組和模型組給予等體積的超純水1個(gè)月;HE染色觀察慢阻肺大鼠肺組織變化;TUNEL法檢測(cè)凋亡細(xì)胞并計(jì)算凋亡指數(shù)(AI);Elisa法檢測(cè)肺泡灌洗液(BALF)中血管內(nèi)皮生長(zhǎng)因子(VEGF)水平;Western blotting法檢測(cè)肺組織中VEGF和血管內(nèi)皮生長(zhǎng)因子受體2(VEGFR2)的蛋白表達(dá)。結(jié)果與對(duì)照組比較,模型組大鼠肺組織形態(tài)病變明顯;AI顯著升高;BALF中VEGF、肺組織中VEGF和VEGFR2水平均顯著下降。與模型組比較,潤(rùn)肺1月和3月組肺組織形態(tài)明顯好轉(zhuǎn);AI顯著下降;BALF中VEGF、肺組織中VEGF和VEGFR2水平顯著升高;且潤(rùn)肺3月組效果好于1月組。結(jié)論潤(rùn)肺止嗽丸可以改善慢阻肺細(xì)胞凋亡,機(jī)制可能與上調(diào)VEGF和VEGFR2相關(guān)。

[Abstract]
Objective To study the effect of Runfei Zhisou Pills (RZP) on cell apoptosis in rat model of chronic obstructive pulmonary disease (COPD) and its mechanism. Methods Totally 32 male Wistar rats were randomly divided into control group, model group, 1-month RZP group, and 3-month RZP group. Rats in 1-month and 3-month RZP group, received RZP of 0.9 g/kg once daily for 1 and 3 months respectively, and rats in control and model groups received water with equal volume for 1 month. The rat COPD model was established using tobacco smoke combined with intratracheal instillation of LPS. Morphological changes of lung tissue in COPD rats were observed with HE staining under light microscope. The VEGF levels in bronchoalveolar lavage fluid (BALF) were detected by ELISA. The VEGF and VEGFR2 protein expression in lung tissues was measured by Western blotting. Cell apoptosis in lung tissues was detected with TUNEL method, and apoptosis index (AI) was counted. Results Compared with control group, the pathological changes of lung tissue in model group were obvious, the AI significantly increased, VEGF in BALF and VEGF and VEGFR2 in lung tissue decreased significantly. Compared with model group, morphological improvement of lung tissue was obvious, the AI significantly decreased, and VEGF in BALF, VEGF and VEGFR2 in lung tissue increased significantly in 1-month and 3-month RZP groups. Conclusion RZP can inhibit cell apoptosis of COPD, and the mechanism may be related with up-regulation of VEGF and VEGFR2.
