[摘要]
目的 觀察白頭翁湯對鏈脲佐菌素(STZ)誘導(dǎo)的I型糖尿病小鼠肝損傷的治療效果。方法 單次ip STZ(180 mg/kg)建立1型糖尿病小鼠模型,對照組給予等量的檸檬酸緩沖液。挑選血糖值≥ 15 mmol/L的小鼠隨機(jī)分為模型組、二甲雙胍組及白頭翁湯低、中、高劑量(生藥2.5、5.0、10.0 g/kg)組,對照和模型組給予等體積生理鹽水,連續(xù)ig給藥4周。末次給藥24 h后,稱各組小鼠體質(zhì)量,測空腹血糖;試劑盒法檢測血清中丙氨酸轉(zhuǎn)氨酶(ALT)、天冬氨酸轉(zhuǎn)氨酶(AST)、堿性磷酸酶(ALP)、腫瘤壞死因子-α(TNF-α)、白介素-6(IL-6)水平,檢測血清及肝組織中丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽過氧化物酶(GSH-Px)水平,HE染色觀察肝組織病理組織學(xué)損傷。結(jié)果 給藥4周后,與模型組比較,白頭翁湯中、高劑量組動物體質(zhì)量顯著升高、空腹血糖值顯著降低(P<0.05、0.01);中、高劑量組血清ALT、AST、ALP、TNF-α、IL-6水平顯著下降(P<0.05、0.01、0.001);中、高劑量顯著降低血清及肝組織中MDA含量,高劑量顯著提升血清及肝組織中SOD活力,中、高劑量顯著增加血清GSH-Px水平,高劑量顯著增加肝組織GSH-Px水平(P<0.05、0.01)。HE染色顯示白頭翁湯可以改善肝臟病理組織學(xué)損傷。結(jié)論 白頭翁湯可以有效改善STZ誘導(dǎo)的糖尿病肝損傷,機(jī)制與減輕脂質(zhì)過氧化、增強(qiáng)機(jī)體清除自由基能力以及抗炎作用有關(guān)。

[Abstract]
Objective To investigat the protective effect of Pulsatillae Decoction on streptozocin (STZ) induced Type I diabetes. Methods The diabetic model was induced with a single dose of 180 mg/kg STZ. The fasting blood glucose over 15 mmol/L was considered as diabetes. The diabetic mice were randomly divided into five groups:model group, metformin group, Pulsatillae Decoction low, medium, and high dose groups. The mice were ig administrated with the drugs for continuous four weeks, then the body weight, fast blood glucose, the levels of ALT, AST, ALP, TNF-α, and IL-6 in serum were detected. The levels of MDA, SOD, GSH-Px in serum and hepatic tissues were also examined. The histopathological changes were observed by H&E staining. Results After a four week treatment, Pulsatillae Decoction medium and high dose groups significantly increased the body weight, decreased the fast blood glucose (P<0.05 and 0.01), reduced the levels of ALT, AST, ALP, TNF-α, and IL-6 (P<0.05, 0.01, and 0.001), attenuated the levels of MDA, SOD, and GSH-Px in serum and hepatic tissues (P<0.05 and 0.01). The results of HE staining also showed that Pulsatillae Decoction was able to relive the histopathological alteration. Conclusion Pulsatillae Decoction could ameliorate the STZ-induced diabetic hepatic injury, possibly via the scavenging Oxygen free radical, anti-oxidative, and anti-inflammatory activities.
