[摘要]
目的 探討坤泰膠囊對(duì)不同治療方法下卵巢早衰大鼠卵巢功能和生殖功能的影響。方法 將84只SD雌性大鼠隨機(jī)分為卵巢功能組和生殖功能組兩個(gè)大組,分別觀察卵巢功能和生殖功能,每個(gè)大組分為空白組、模型組和坤泰膠囊高、中、低劑量組及結(jié)合雌激素片組,采用ig雷公藤多苷片(75 mg/kg,14 d)建立卵巢早衰模型,造模結(jié)束后,分別用坤泰膠囊高、中、低劑量(1.2、0.6、0.3 g/kg,36 d)及結(jié)合雌激素片(0.075 mg/kg,36 d)進(jìn)行治療。治療結(jié)束后,卵巢功能組所有大鼠用酶聯(lián)免疫吸附試驗(yàn)(ELISA)法檢測(cè)大鼠血清性激素水平的變化,用實(shí)時(shí)熒光定量PCR(qRT-PCR)法檢測(cè)大鼠卵巢組織相關(guān)因子Fas、FasL mRNA的表達(dá)水平的變化。將生殖功能組所有雌性大鼠與12只SD雄性大鼠進(jìn)行合籠,觀察記錄孕鼠一般情況及子鼠的生長(zhǎng)發(fā)育情況,觀察各組雌性大鼠的生殖能力。結(jié)果 治療組與模型組比較,隨著治療天數(shù)的增加,大鼠的卵巢功能和生殖功能均明顯改善,血清卵泡生成激素(FSH)、黃體生成激素(LH)水平明顯降低,雌二醇(E2)、抗穆勒管激素(AMH)水平明顯升高,F(xiàn)asmRNA表達(dá)明顯升高、FasLmRNA表達(dá)明顯降低,差異均有統(tǒng)計(jì)學(xué)意義(P<0.05);孕鼠攝食量及體質(zhì)量均增長(zhǎng)較快,差異有統(tǒng)計(jì)學(xué)意義(P<0.05)。大鼠產(chǎn)仔總數(shù)多,受孕率、妊娠率均較高,無明顯子代小鼠畸形率,差異有統(tǒng)計(jì)學(xué)意義(P<0.05)。結(jié)論 坤泰膠囊能夠改善卵巢早衰大鼠的卵巢功能和生殖功能,可修復(fù)育齡期大鼠卵巢功能病理性損傷,有效調(diào)節(jié)卵巢早衰大鼠血清性激素水平,上調(diào)FasmRNA表達(dá),下調(diào)FasLmRNA表達(dá),提高卵巢早衰大鼠受孕率及妊娠率,對(duì)子代小鼠無明顯的致畸性。

[Abstract]
Objective To investigate the effects of Kuntai Capsule on ovarian function and reproductive function in rats with premature ovarian failure under different treatments. Methods 84 SD female rats were randomly divided into two groups:ovarian function group and reproductive function group. The ovarian function and reproductive function were observed. Each group was divided into blank group, model group and Kuntai capsule high, medium, low dose group and conjugated estrogen tablet group. The premature ovarian failure model was established by Ig Tripterygium glycoside tablet (75 mg/kg, 14 days). After the model was established, Kuntai capsule high dose group was used. Medium and low doses (1.2, 0.6, 0.3 g/kg, 36 days) and conjugated estrogen tablets (0.075 mg/kg, 36 days) were administered. After treatment, all rats in ovarian function group were tested for the changes of serum sex hormone levels by ELISA, and the expression levels of Fas and FasL in ovarian tissue were detected by real-time fluorescence quantitative PCR (qRT-PCR). All female rats in reproductive function group were caged with 12 SD male rats. The general situation of pregnant rats and the growth and development of offspring were observed and recorded, and the reproductive ability of female rats in each group was observed. Results Compared with the model group, the ovarian function and reproductive function of rats were improved significantly with the increase of treatment days. The levels of serum follicle-forming hormone (FSH) and luteinizing hormone (LH) were significantly decreased, the levels of estradiol (E2) and anti-Mullerian hormone (AMH) were significantly increased, the expression of Fas gene was significantly increased, and the expression of FasL gene was significantly decreased in the treatment group (P The intake and body mass of pregnant rats increased rapidly, and the difference was statistically significant (P<0.05). The total number of offspring of rats was large, the pregnancy rate and pregnancy rate were high, and there was no obvious deformity rate in offspring of mice, the difference was statistically significant (P<0.05). Conclusion Kuntai Capsule can improve the ovarian function and reproductive function of premature ovarian failure rats, repair the pathological damage of ovarian function in reproductive age rats, effectively regulate the level of serum sex hormones, up-regulate the expression of Fas gene, down-regulate the expression of FasL gene, improve the pregnancy rate and pregnancy rate of premature ovarian failure rats, and has no obvious teratogenicity to offspring mice.
