50);ELISA法檢測(cè)IC50的青霉素對(duì)血小板中蛋白激酶A(Protein kinase A,PKA)、β血小板球蛋白(β-thromboglobulin,β-TG)、超氧化物歧化酶2(Superoxide dismutase 2,SOD2)、血小板第4因子(Platelet factor 4,PF4)和血小板內(nèi)皮細(xì)胞黏附分子-1(Plateletendothelial cell adhesion molecule-1,PECAM-1)含量的影響。結(jié)果 隨著青霉素濃度的增大,血小板數(shù)目逐漸減少,血小板存活率逐漸降低,IC50值為985.68 U/mL。與對(duì)照組比較,青霉素組的PKA、β-TG、SOD2、PF4和PECAM-1的含量顯著降低,差異具有統(tǒng)計(jì)學(xué)意義(P<0.05)。結(jié)論 青霉素可能通過(guò)降低PKA、SOD2、β-TG、PF4和PECAM-1含量,進(jìn)而顯著降低血小板存活率,抑制血小板活性及其聚集功能。;Objective To investigate the effects of penicillin on activity, longevity and adhesion-related functions of platelet. Methods Peripheral platelets of C57BL/6J mice were isolated. Platelets were treated with 10, 50, 100, 500, 1 000, 5 000 U/mL penicillin for 24 h. MTT was used to detect platelet activity and IC50 values were calculated. The study was divided into control group and penicillin group. The contents of protein kinase A (PKA), β-thromboglobulin (β-TG), superoxide dismutase 2 (SOD2), Platelet factor 4 (PF4) and platelet endothelial cell adhesion molecule-1 (PECAM-1) in platelets were detected by ELISA. Results With the increase of penicillin concentration, the number of platelets decreased gradually, and the platelet survival rate gradually decreased. The calculated IC 50 value is 985.68 U/mL. Compared with the control group, the contents of PKA, β-TG, SOD2, PF4 and PECAM-1 in the penicillin group were significantly lower, and the difference was statistically significant (P<0.05). Conclusion Penicillin may significantly reduce platelet survival rate, platelet activity and aggregation function by reducing the contents of PKA, SOD2, β-TG, PF4 and PECAM-1 in platelets."/>
