[摘要]
目的 研究治咳川貝枇杷滴丸(CBPP)的祛痰、鎮(zhèn)咳作用,以及對(duì)肺組織Muc5AC表達(dá)的影響,明確其藥效特點(diǎn),為中藥臨床定位提供實(shí)驗(yàn)依據(jù)。方法 采用雄性SPF級(jí)ICR小鼠,分別為對(duì)照(生理鹽水)組、鹽酸氨溴索口服液(陽(yáng)性藥,臨床等效劑量11.7 mg/kg)組和CBPP低、中、高劑量(70.2、140.4、280.8 mg/kg)組,采用氣管酚紅排泌實(shí)驗(yàn)結(jié)合分光光度計(jì)法探究CBPP的祛痰藥效;利用氨水引咳實(shí)驗(yàn)觀察CBPP高劑量組的鎮(zhèn)咳作用;采用實(shí)時(shí)熒光定量PCR(qRT-PCR)法檢測(cè)CBPP高劑量組對(duì)肺組織中Muc5AC mRNA表達(dá)的影響。家兔隨機(jī)分為對(duì)照組、鹽酸氨溴索口服液(10.5 mg/mL)組、CBPP(252 mg/mL)組,利用離體氣管纖毛運(yùn)動(dòng)實(shí)驗(yàn)考察CBPP對(duì)氣管纖毛運(yùn)動(dòng)的影響。結(jié)果 酚紅排泌實(shí)驗(yàn)顯示,與對(duì)照組比較,陽(yáng)性藥和CBPP顯著促進(jìn)正常小鼠氣管酚紅排泌量增加(P<0.05、0.01),且CBPP作用呈劑量相關(guān)性;家兔離體氣管纖毛運(yùn)動(dòng)實(shí)驗(yàn)顯示,與對(duì)照組比較,陽(yáng)性藥和CBPP顯著促進(jìn)纖毛運(yùn)動(dòng)(P<0.05),進(jìn)而促進(jìn)痰液排出;氨水引咳實(shí)驗(yàn)顯示,與對(duì)照組比較,CBPP顯著延長(zhǎng)小鼠的咳嗽潛伏期、減少咳嗽次數(shù)(P<0.05);qRT-PCR結(jié)果顯示,與對(duì)照組比較,CBPP顯著降低肺組織中Muc5AC的mRNA表達(dá)(P<0.05)。結(jié)論 CBPP具有明顯的祛痰作用,促進(jìn)呼吸道腺體的分泌,并促進(jìn)氣管纖毛運(yùn)動(dòng),降低Muc5AC的含量,促進(jìn)痰液排出,并且對(duì)氨水刺激引起的咳嗽有一定的鎮(zhèn)咳作用。

Objective To study the expectorant and antitussive effects of Zhike Chuanbei Pipa Dropping Pills (CBPP), and its effect on the expression of MUC5AC in lung tissue, clarify their pharmacodynamics characteristics, and provide experimental basis for the clinical positioning of this Chinese patent medicine. Methods Male SPF ICR mice were randomly divided into control group, positive drug group (ambroxol hydrochloride oral solution, 11.7 mg/kg), and CBPP low, medium and high-dose (70.2, 140.4, 280.8 mg/kg) group. Observe the expectorant effects of CBPP by phenol red expectorant experiment combined spectrophotometer method in normal mice. Rabbits were randomly divided intocontrol group, positive drug group (ambroxol hydrochloride oral solution,10.5 mg/mL) and CBPP (252 mg/mL) group. The rabbit in vitro tracheal ciliary exercise experiment was used to examine the effect of CBPP on tracheal cilia movement. The antitussive effect of CBPP of high dose by using normal mice with ammonia water induced cough experiment. Detect the content of Muc5AC mRNA in lung tissue by qRT-PCR in control and CBPP high dose group. Results Phenol red excretion experiments showed that compared with control group, the positive drug and CBPP promoted the increase of tracheal phenol red excretion in mice (P<0.05 or 0.01), and promoted the secretion of respiratory tract glands in mice, and CBPP had a certain dose-dependent effect. Rabbit in vitro tracheal ciliary exercise experiment showed treatment of positive drug and high dose of CBPP promoted cilia movement (P<0.05), and then promoted sputum excretion. Ammonia water-induced cough experiment showed that compared with control group, CBPP high-dose group prolonged the cough latency period of mice and reduced the number of coughs (P<0.05). qRT-PCR results showed that compared with control group, CBPP administration reduced the content of Muc5AC mRNA in lung tissue (P<0.05). Conclusion CBPP has obvious expectorant effect, promotes the secretion of respiratory tract glands, and promotes the movement of tracheal cilia, reduces the content of Muc5AC, promotes sputum discharge, and has a certain antitussive effect on cough caused by ammonia stimulation.
