[摘要]
目的 運用網(wǎng)絡(luò)藥理學(xué)的方法挖掘中藥復(fù)方急支糖漿治療咳嗽的有效成分、關(guān)鍵靶點以及潛在作用通路。方法 分別利用中藥系統(tǒng)藥理學(xué)數(shù)據(jù)庫與分析平臺(TCMSP)和Genecards數(shù)據(jù)庫篩選急支糖漿的活性成分作用靶點和咳嗽的疾病靶點,兩者靶點取交集后利用Cytoscape軟件構(gòu)建成分-靶點網(wǎng)絡(luò),String數(shù)據(jù)庫構(gòu)建蛋白質(zhì)相互作用(PPI)網(wǎng)絡(luò),拓撲分析獲取關(guān)鍵靶點和成分。通過David數(shù)據(jù)庫進行基因本體論(GO)生物學(xué)功能富集分析及京都基因和基因組百科全書(KEGG)信號通路富集分析。通過分子對接技術(shù)進一步佐證網(wǎng)絡(luò)藥理學(xué)結(jié)果的可靠性,通過小鼠咳嗽模型進行實驗驗證。結(jié)果 急支糖漿中共篩選出活性成分163種,可干預(yù)咳嗽相關(guān)靶點154個,GO富集586個,KEGG通路119條。其中,槲皮素、山柰酚、β-谷甾醇等可能為急支糖漿治療咳嗽的關(guān)鍵成分,CXCL8、MAPK8、STAT3、TNF等為潛在靶點,GO富集分析的生物功能涉及到細胞增殖、血管生成、炎癥反應(yīng)等,KEGG通路富集分析涉及到TNF、PI3K-Akt、FoxO、HIF-1等信號通路。超過90%的分子對接結(jié)果均具有一定的結(jié)合活性。使用流式細胞術(shù)、實時熒光定量PCR(qRT-PCR)、酶聯(lián)免疫吸附(ELISA)的方法驗證了急支糖漿對咳嗽小鼠淋巴細胞分化、炎癥因子和部分關(guān)鍵靶點表達的影響。結(jié)論 急支糖漿治療咳嗽的作用機制是通過多種活性成分作用于多靶點、多通路產(chǎn)生了調(diào)控炎癥過程、調(diào)節(jié)免疫功能、促進細胞增殖、修復(fù)受損機體等綜合效果,該結(jié)果為進一步研究急支糖漿有效成分和作用機制奠定基礎(chǔ)。

Objective To explore effective ingredients, key targets, and potential pathways of traditional Chinese medicine compound Jizhi Syrup for treatment of cough by method of network pharmacology. Methods TCMSP and genecards databases were used to screen active component action targets of Jizhi Syrup and disease targets of cough. After intersection of two targets, the component target network was constructed by Cytoscape software, and the protein-protein interaction network (PPI) was constructed by string database. The key targets and components were obtained by topological analysis. Gene Ontology (GO) biological function enrichment analysis and Kyoto encyclopedia of genes and genomes (KEGG) signal pathway enrichment analysis were carried out through David database. The reliability of network pharmacological results was further confirmed by molecular docking technology, and verified by mouse cough model. Results A total of 163 active ingredients were selected from Jizhi Syrup, which could intervene 154 cough related targets, 586 GO enrichment and 119 KEGG pathways. Quercetin, kaempferol β-sitosterol may be the key components of Jizhi Syrup in treatment of cough. CXCL8, MAPK8, STAT3 and TNF are potential targets. The biological functions of GO enrichment analysis involve cell proliferation, angiogenesis and inflammatory response. KEGG pathway enrichment analysis involves TNF, PI3K-Akt, FOXO, HIF-1 and other signal pathways. More than 90% of the molecular docking results have certain binding activity. The effects of Jizhi Syrup on lymphocyte differentiation, inflammatory factors and the expression of some key targets in cough mice were verified by flow cytometry, real-time fluorescence quantitative PCR (qRT-PCR) and enzyme-linked immunosorbent assay (ELISA). Conclusion The mechanism of Jizhi Syrup in treatment of cough is that multiple active components act on multiple targets and pathways, resulting in comprehensive effects of regulating inflammatory process, regulating immune function, promoting cell proliferation and repairing damaged body. The results lay a foundation for the further study of the effective components and mechanism of Jizhi Syrup.
