阿爾茨海默?。ˋD)是一種復(fù)雜且與年齡相關(guān)的進(jìn)行性神經(jīng)退行性疾病,是最常見的癡呆癥類型,其特征是認(rèn)知功能、語言和行為缺陷逐漸惡化。β-淀粉樣蛋白(Aβ)異常沉積、神經(jīng)原纖維纏結(jié)(NFT)、低水平乙酰膽堿、鈣離子穩(wěn)態(tài)失衡、氧化應(yīng)激、神經(jīng)炎癥等被認(rèn)為在AD的發(fā)病機(jī)制中起著關(guān)鍵作用。核因子E2相關(guān)因子2(Nrf2)是一種重要的轉(zhuǎn)錄調(diào)節(jié)因子,在氧化還原穩(wěn)態(tài)、蛋白質(zhì)降解、DNA修復(fù)以及調(diào)節(jié)外源代謝和免疫反應(yīng)中發(fā)揮多種作用,已成為炎癥、腫瘤治療等領(lǐng)域研究的主要靶點,在AD的防治方面已被廣泛關(guān)注。中藥天然產(chǎn)物在AD防治過程中發(fā)揮著不可替代的作用?;诖?,對Nrf2與AD的關(guān)系及中藥天然產(chǎn)物調(diào)控Nrf2信號通路防治AD的機(jī)制進(jìn)行總結(jié)歸納,以期為AD的治療及新藥開發(fā)提供參考。

Alzheimer's disease (AD) is a complex and age-related progressive neurodegenerative disorder, the most common form of dementia, characterized by the progressive deterioration of cognitive function, language, and behavioral deficits. Abnormal deposition of beta amyloid protein (Aβ), neurofibrillary tangles (NFT), low levels of acetylcholine, Calcium ion steady-state imbalance, oxidative stress, and neuroinflammation are believed to play key roles in the pathogenesis of AD. Nrf2 is a significant transcriptional regulator that plays multiple roles in redox homeostasis, protein degradation, DNA repair, regulation of exogenous metabolism, and immune response. Nrf2 has emerged as a major target in the fields of inflammation and tumor therapy and has received extensive attention in the prevention and treatment of AD. The natural products of Chinese medicine play an irreplaceable role in the prevention and treatment of AD. Based on this, summarize and generalize the relationship between Nrf2 and AD, as well as the mechanism of regulating the Nrf2 signaling pathway by natural Chinese medicine products to prevent and treat AD, in order to provide reference for the treatment of AD and the development of new drugs.
