[摘要]
目的 運(yùn)用中藥整合藥理學(xué)計(jì)算平臺(TCMIP),探索巴戟天治療抑郁癥的作用機(jī)制。方法 借助TCMIP,檢索巴戟天所含的化學(xué)成分信息;通過網(wǎng)絡(luò)藥理學(xué)平臺,以"節(jié)點(diǎn)連接度"的2倍中位數(shù)為卡值,選取中藥靶標(biāo)-疾病基因互作網(wǎng)絡(luò)的核心節(jié)點(diǎn);在此基礎(chǔ)上,計(jì)算"節(jié)點(diǎn)連接度""節(jié)點(diǎn)緊密度"和"節(jié)點(diǎn)介度"等3個(gè)拓?fù)浣Y(jié)構(gòu)特征值,篩選出巴戟天治療抑郁癥的核心靶標(biāo);基于基因本體數(shù)據(jù)庫(GO)和KEGG通路數(shù)據(jù)庫,確定藥物靶標(biāo)基因所編碼蛋白質(zhì)的分子功能、細(xì)胞內(nèi)定位及其所參與的生物學(xué)反應(yīng)和通路,再進(jìn)一步構(gòu)建"中藥-成分-靶標(biāo)-通路"多維關(guān)系網(wǎng)絡(luò)。結(jié)果 測得巴戟天所含活性成分19個(gè),主要包括蒽醌類、多糖類、環(huán)烯醚萜苷類及其他成分;篩選出巴戟天治療抑郁癥的3個(gè)核心靶標(biāo),包括5-羥色胺受體2A、2B、2C(HTR2A、HTR2B、HTR2c);結(jié)合富集計(jì)算,預(yù)測巴戟天治療抑郁癥可能與氧化磷酸化、能量代謝、鈣信號通路、心肌收縮、內(nèi)分泌系統(tǒng)、循環(huán)系統(tǒng)、神經(jīng)系統(tǒng)、感覺系統(tǒng)、細(xì)胞通訊等通路有關(guān)。結(jié)論 借助TCMIP,發(fā)現(xiàn)巴戟天對抑郁癥的干預(yù)作用可能與氧化磷酸化、能量代謝、鈣信號通路、內(nèi)分泌系統(tǒng)、循環(huán)系統(tǒng)、神經(jīng)系統(tǒng)、感覺系統(tǒng)、細(xì)胞通訊等通路有關(guān)。

[Abstract]
Objective The integrated pharmacology platform was used to explore the mechanism of morinda in the treatment of depression. Methods With TCMIP, the chemical composition information of M. officinalis was retrieved. Taking the median of 2 times of "node connectivity" as the card value, the core node of the network of Chinese medicine target-disease gene interaction was selected. On this basis, three topological feature values such as "node connectivity" "node tightness" and "node interface" were calculated to screen out core targets for treating depression by M. officinalis. Based on GO and KEGG pathway databases, molecular functions, intracellular localization of proteins encoded by drug target genes and biological responses and pathways involved are determined. The multi-dimensional relationship network of "TCM-component-target-pathway" was further constructed. Results There were 19 active compounds, including anthraquinone, polysaccharides, iridoid glycosides and other compounds.Through the network pharmacology platform, three core targets for treating depression by halberd, including 5-hydroxytryptamine receptor 2A (HTR2A), 5-hydroxytryptamine receptor 2B (HTR2B) and 5-hydroxytryptamine receptor 2c (HTR2c). Combined with enrichment calculation, it was predicted that the treatment of depression by morinda may be related through such pathways as oxidative phosphorylation, energy metabolism, calcium signaling pathway, myocardial contraction, endocrine system, circulatory system, nervous system, sensory system and cell communication.
