[摘要]
目的 研究不同培養(yǎng)條件對鐵皮石斛類原球莖在生物反應(yīng)器中培養(yǎng)的影響。方法 采用接種量、通氣量、蔗糖濃度、光照強度等單因素試驗設(shè)計,烘干法測定生物量,苯酚-硫酸法測定多糖量,數(shù)據(jù)采用SPSS16.0軟件分析。結(jié)果 在生物反應(yīng)器培養(yǎng)條件下,利于鐵皮石斛類原球莖增殖和多糖積累的培養(yǎng)基組分為:1/2 MS基本培養(yǎng)基添加1.0 mg/L NAA、5%椰乳、3%蔗糖,pH值為6.0。接種量為40 g/L,采用孔徑為15 μm的多孔噴頭,通氣量用1.0 L/min和0. 5 L/min交替使用以及光照強度為2 000 lx等培養(yǎng)條件有效地提高類原球莖增殖系數(shù)和多糖量。結(jié)論 不同的培養(yǎng)條件對鐵皮石斛類原球莖的生長、多糖的變化有顯著影響,適宜的培養(yǎng)條件對鐵皮石斛類原球莖的生物量和主要藥用成分的生產(chǎn)具有重要的實踐意義。

[Abstract]
Objective To explore the effect of different culture conditions on protocorm-like bodies (PLBs ) of Dendrobium candidum in bioreactor culture. Methods Inoculation quantity, ventilation, sucrose concentration, and light intensity were taken to carriy out the single factor experiment design. Biomass was determined by drying method, polysaccharide content by phenol-sulfuric acid method and SPSS16.0 software was used for data analysis. Results Under the bioreactor culture, the nutrient media components for the proliferation of PLBs of D. candidum and accumulation of polysaccharide were1/2 MS medium supplemented with 1.0 mg/L NAA, 5% coconutmilk, and 3% sucrose, 6.0 pH, was benefit for the proliferation and polysaccharide accumulation of the PLBs. The culture conditions such as inoculation with 40 g/L,, 15 μm pore porous nozzle, aeration volumes by 1.0 and 0.5 L/min used alternately, and photon flux of 2 000 lx could improve the proliferationg coefficient and polysaccharide contents of PLBs. Conclusion Different culture conditions have the significant effect on the growth of PLBs and the accumulation of polysaccharide. Suitable culture conditions have the very important practical significance to the biomass of PLBs and main medicinal ingredient production of PLBs.
