[摘要]
目的 研究安息香Benzoinum化學(xué)成分以及體外抑制腫瘤增殖活性。方法 采用MCI gel CHP-20P、正相硅膠、ODS和Sephadex LH-20柱色譜以及半制備型高效液相等色譜方法進行分離和純化,通過紅外、核磁共振、質(zhì)譜等技術(shù)鑒定化合物的結(jié)構(gòu)。CCK-8法評價分離得到的化合物體外抑制腫瘤增殖的活性。結(jié)果 從安息香醋酸乙酯萃取部位中分離得到3個化合物,分別鑒定為1,3-二苯甲酸酯-2-(羥基(4-羥基-3-甲氧基苯基)甲基)丙烷(1)、5-乙?;?7-甲氧基-2-(3,4-亞甲二氧基苯基)苯駢呋喃(2)和5-甲?;?7-甲氧基-2-(3,4-亞甲二氧基苯基)苯駢呋喃(3)?;衔?b>2和3對人肝癌HepG2細胞增殖有抑制作用,半數(shù)抑制濃度(half maximal inhibitory concentration,IC50)值分別為41.2、52.6μmol/L。結(jié)論 化合物1和2為新化合物,命名為白花樹醇酯(1)和白花樹醇酮(2);化合物3為新的天然產(chǎn)物;其中化合物2和3對人肝癌HepG2細胞增殖表現(xiàn)出一定的細胞毒活性。

[Abstract]
Objective To study the chemical constituents from Benzoinum and their antitumor activities in vitro. Methods The compounds were isolated and purified by MCI gel CHP-20P, silica gel, ODS, Sephadex LH-20 column chromatography and semi-preparative HPLC. Their structures were elucidated by spectroscopic analysis including IR, NMR and MS. CCK-8 assay was used to evaluate the inhibitory activity of the isolated compounds on tumor proliferation in vitro. Results A total of three compounds, 1,3-dibenzoate-2-(hydroxy(4-hydroxy-3-methoxyphenyl) methyl)propane (1), 5-acetyl-7-methoxy-2-(3,4-methylenedioxyphenyl) benzofuran (2), and 5-formyl-7-methoxy-2-(3,4-methylenedioxyphenyl)benzofuran (3), were isolated from the ethyl acetate extraction site of Benzoinum. Compounds 2 and 3 exhibited the inhibitory effects on HepG-2 cells with half maximal inhibitory concentration (IC50) values of 41.2, 52.6 μmol·L−1, respectively. Conclusion Compounds 1 and 2 are new compounds named styratonate (1) and styratonone (2), and compound 3 is a new natural product, among which compounds 2 and 3 exhibit certain cytotoxic activities on the proliferation of HepG2 cells.
