[摘要]
目的 優(yōu)選建曲發(fā)酵工藝,為建曲發(fā)酵的規(guī)范化生產(chǎn)提供參考依據(jù)。方法 以橙皮苷、柚皮素、橙皮素、醇溶性浸出物含量、糖化酶活力及外觀性狀為評(píng)價(jià)指標(biāo),對(duì)炮制過(guò)程的發(fā)酵時(shí)間、發(fā)酵溫度、相對(duì)濕度(relative humidity,RH)進(jìn)行考察,通過(guò)AHP-CRITIC綜合加權(quán)法確定各評(píng)價(jià)指標(biāo)的權(quán)重系數(shù),聯(lián)合單因素實(shí)驗(yàn)與Box-Behnken設(shè)計(jì)-響應(yīng)面法,以綜合評(píng)分作為評(píng)價(jià)指標(biāo),優(yōu)選最佳發(fā)酵工藝參數(shù)。結(jié)果AHP-CRITIC綜合賦權(quán)法確定的橙皮苷、柚皮素、橙皮素、醇溶性浸出物含量及糖化酶活力和外觀性狀的權(quán)重系數(shù)分別為0.070 2、0.107 1、0.078 0、0.058 5、0.235 4、0.450 8;優(yōu)選的建曲發(fā)酵工藝為將處方中各中藥細(xì)粉、麥麩與面粉稀糊揉合均勻并壓制成塊后,置發(fā)酵箱內(nèi),在發(fā)酵溫度35 ℃、RH 80%條件下連續(xù)發(fā)酵23 h。平行3次的驗(yàn)證試驗(yàn)綜合評(píng)分均值為87.889,與預(yù)測(cè)值的相對(duì)偏差為1.07%,且各質(zhì)量指標(biāo)的RSD均小于3.0%。結(jié)論 采用響應(yīng)面法建立的模型科學(xué)準(zhǔn)確,優(yōu)選的建曲發(fā)酵工藝穩(wěn)定可行,可為以建曲為代表的發(fā)酵類中藥規(guī)范化生產(chǎn)和質(zhì)量標(biāo)準(zhǔn)提高提供參考依據(jù)。

[Abstract]
Objective To optimize the fermentation process of Jianqu and provide reference for the standardized production of Jianqu fermentation. Methods Using the contents of hesperidin, naringenin, hesperetin and alcohol soluble extract, glucoamylase activity and appearance characteristics. as evaluation indexes, the fermentation time, fermentation temperature and relative humidity (RH) during processing were investigated, and the weight coefficients of each index were determined by AHP-CRITIC hybrid weighting method. Combined with single factor experiment and Box-Behnken design-response surface method, the comprehensive score was used as the evaluation index to optimize the fermentation process parameters. Results The weight coefficients of hesperidin, naringenin, hesperetin and alcohol soluble extract, glucoamylase activity and appearance traits determined by AHP-CRITIC comprehensive weighting method were 0.070 2, 0.107 1, 0.078 0, 0.058 5, 0.235 4 and 0.450 8, respectively. The optimal fermentation process of Jianqu was as follows: the fine powder of traditional Chinese medicine, wheat bran and flour paste in the prescription were evenly kneaded and pressed into blocks, and then placed in a fermentation box for continuous fermentation for 23 h at fermentation temperature of 35 ℃ and fermentation humidity of RH 80%. The average comprehensive score of three parallel verification tests was 87.889, the relative deviation from the predicted value was 1.07%, and the RSD of each quality index was less than 3.0%. Conclusion The model established by response surface method is scientific and accurate, and the optimized fermentation process of Jianqu is stable and feasible, which can provide reference for standardized production and quality standard improvement of fermented Chinese medicines represented by Jianqu.
